Sammendrag
Fenylpropanoider er plante sekundærmetabolitter og har i de siste årene fått oppmerksomhet da de har vist forskjellige biologiske effekter. Målet for dette master prosjektet var å danne et kjemisk bibliotek av fenylpropanoid forbindelser med følgende fellestrekk: en benzenring og en (E)-α,β-umettet-ester eller et (E)-α,β-umettet-amid, ved hjelp av tradisjonell syntese. Det ble brukt trekomponentsyntese med et aldehyd, Bestmann-ylid [(trifenylfosforanyliden)keten] (1) og en alkohol eller et amin til syntetisering av forbindelsene. Den foreslåtte syntesen av forbindelsene er en kort vei som Bestmann-ylid (1) inngår for å koble sammen alkohol- eller aminkjeden med karbonylkarbonet i aldehydet og resulterer i dannelse av (E)-α,β-umettet-ester eller -amid. Den første planen for syntetisering av forbindelsene var bruk av termisk varmetilførsel til en kolbe gjennom et oljebad. Denne tilnærmingen resulterte ikke i ønskede reaksjonsutbytter, dermed ble mikrobølgeovn og trykkrør benyttet for å oppnå ønskede resultater.
Synteserute I Synteserute til dannelse av fenylpropanoid forbindelserFør igangsettelse av trekomponentsyntesen, måtte noen av aldehydene beskyttes da de har en eller to fenoliske hydroksylgruppe. Det ble brukt silylbeskyttelsesgruppene TBS-Cl og TIPS-Cl da disse er mye brukt i de siste årene til beskyttelse av hydroksyl. Etter dannelse av ønskede forbindelser gjennom trekomponentsyntese, ble forbindelsene med silylbeskyttelsesgrupper avbeskyttet ved hjelp av reagens tetra-n-butylammoniumfluorid (TBAF). Avbeskyttelsesreaksjonen med (E)-α,β-umettet-ester var vellykket, men det ble ikke forsøkt med (E)-α,β-umettet-amid da (E)-α,β-umettet-amid-forbindelser med beskyttelsesgruppe ikke ble isolert. Den utvalgte synteseruten har dannet flere biprodukter, noe som har gjort separasjonsmetoden krevende. Det ble syntetisert 30 forbindelser, men kun 18 forbindelser er isolert og identifisert. Fenylpropanoids are plant secondary metabolites and have gained attention in recent years as they have shown various biological effects. The aim of this master's project was to form a chemical library of fenylpropanoid compounds with the following common features: a benzene ring and an (E)-α,β-unsaturated ester or an (E)-α,β-unsaturated amide, using traditional synthesis. Three-component synthesis with an aldehyde, Bestmann ylide [(trifenylphosphoranylidene)ketene] and an alcohol or an amine was used to synthesize the compounds. The proposed synthesis of the compounds is a short route involving Bestmann ylide to connect the alcohol or amine chain with the carbonyl carbon of the aldehyde and results in the formation of (E)-α,β-unsaturated ester or amide. The first plan for synthesizing the compounds was the use of thermal heat input to a flask through an oil bath. This approach did not result in the desired reaction yield, therefore microwave heating and pressure tubes were used to achieve the desired result.
Scheme I Synthesis route for the formation of fenylpropanoid compoundsBefore starting the three-component synthesis, some of the aldehydes had to be protected as they have one or two fenolic hydroksyl groups. The silyl protecting groups TBS-Cl and TIPS-Cl were used as these have been widely used in recent years for the protection of hydroksyl. After formation of desired compounds through three-component synthesis, the compounds with silyl protecting groups were deprotected using the reagent tetra-n-butylammonium fluoride (TBAF). The deprotection reaction with (E)-α,β-unsaturated-ester was successful, but it was not attempted with (E)-α,β-unsaturated-amide as (E)-α,β-unsaturated-amide compounds with a protecting group was not isolated. The selected synthesis route formed several by-products, which made the separation method challenging. 30 compounds were synthesized, of which 18 compounds have been isolated and identified.
