Abstract
Målet med denne masteroppgaven er å effektivt syntetisere seks ulike triazolobenzodiazepiner (7) og deres α-hydroksylerte metabolitter (6) fra det korresponderende benzofenonet (1), med fokus på triazolobenzodiazepinene med halogen (klor, brom og fluor) substituenter. En generell fire stegs prosedyre, hvor 1,4-benzodiazepinet (3) også ble syntetisert, ble optimalisert ved å teste ulike løsningsmidler, reagenser og ved å variere reaksjonstiden. Siden prosedyren for å lage 1,4-benzodiapiner er forsket mye på tidligere, var de to siste reaksjonsstegene- tionering av karbonylen og dannelsen av triazolringene av størst interesse. Bruk av tioneringsreagensen Lawessons reagens og direkte dannelse av triazolringene ved bruk av acetohydrazid og 2-hydroxyacetohydrazid ble gjennomført på alle parallellene (a-f). I tillegg til dette ble diclazepam (4a) også syntetisert.
Forbindelse 3a-f, 4a, 6a-f og 7a-f ble syntetisert og karakterisert ved bruk av IR-, 1H-NMR-, and 13C-NMR-spektroskopi. Kromatografisk renhet ble bestemt med HPLC og MS bekreftet molekylformelen. This master thesis aims to effectively synthesize six different triazolobenzodiazepines (7) and their α-hydroxylated metabolites (6), starting with the corresponding benzophenone (1), with the focus on triazolobenzodiazepines having halogen (chloro, bromo and fluoro) substituents. A general four-step procedure in which the 1,4-benzodiazepine (3) was also synthesized, was optimized using different solvents, reagents, and by varying the reaction time. As the procedure making the 1,4-benzodiazepines has been widely researched, the last two steps, thionation of the carbonyl and formation of the triazole rings, were of greater interest. The use of thionating agent Lawesson’s reagent and a direct formation of the triazole ring using acetohydrazide and 2-hydroxyacetohydrazide was applied to all parallels (a-f). In addition to this diclazepam (4a) was synthesised.
Compound 3a-f, 4a, 6a-f and 7a-f were successfully synthesized and characterised using melting points, IR-, 1H-NMR-, and 13C-NMR-spectroscopy. Chromatographic purity was determined by HPLC, and MS confirmed the molecular formula.
