Sammendrag
Bakgrunn: Formålet med denne oppgaven var å starte bruken av plateleseren Tecan SPARK ved instituttet, inkludert å utarbeide en instrumentprotokoll og prosedyrer til instrumentet.
Metode: Til utformingen av instrumentprotokollen og prosedyrene ble det brukt maler for kvalitetssikring av laboratoriearbeid ved NTNU. Innholdet i instrumentprotokollen og prosedyrene ble hentet fra instrumentmanualen og programvaremanualen som ble levert med instrumentet. Prosedyrene og instrumentprotokollen ble utprøvd ved forsøk som hadde til formål å undersøke effekten av ulike innstillinger som ble beskrevet i prosedyrene og instrumentprotokollen.
Resultater: Det ble utarbeidet en instrumentprotokoll, samt prosedyrer for måling av absorbans, fluorescens, kjemiluminescens, celletelling, celleviabilitet og cellekonfluens. Gjennom utprøvingene ble det gjort ulike erfaringer med målemetodene og innstillingene som Tecan SPARK tilbyr. Instrumentprotokollen og prosedyrene var velegnet for utprøving av instrumentet og vil være til stor hjelp ved bruk av instrumentet framover.
Konklusjon: Tecan SPARK har mange målemetoder, hvorav flere har blitt undersøkt og implementert igjennom denne bacheloroppgaven. Prosedyrene og instrumentprotokollen har lagt til rette for måling av absorbans, fluorescens, kjemiluminescens, celletelling, celleviabilitet og cellekonfluens. Background: The purpose of this bachelor thesis is to start the use of Tecan SPARK Plate Reader, including preparation of an instrument protocol and procedures for the use of the instrument.
Method: The preparation of the instrument protocol and the procedures for use of the instrument were based on templates for laboratory quality assurance at NTNU. The contents of the instrument protocol and the procedures were taken from the instrument manual and the software manual delivered with the instrument.
Results: An instrument protocol and procedures for absorbance, fluorescence, chemiluminescence, cell counting, cell viability and cell confluence were established. During testing of the protocol and procedures experiences were made with the different measurement techniques and settings offered by Tecan SPARK. The instrument protocol and procedures proved to be well suitable for operation of Tecan SPARK and will be of great help for the use of the instrument going forward.
Conclusion: Tecan SPARK offers a wide range of measurement techniques, many of which have been explored and implemented by this bachelor thesis. These measurement techniques include absorbance, fluorescence, chemiluminescence, cell counting, cell viability and cell confluence.
