dc.description.abstractUNG2 (uracil DNA-glykosylase) er det dominerende enzymet som fjerner uracil i det humane genomet. Metylert cytosin (5-metylcytosin (5-mC)) utgjør omtrent 1% av det humane DNA, og er med det den vanligste epigenetiske modifikasjonen. Formålet med studien var å undersøke om 5-mC påvirker UNG2-mediert gjenkjenning og fjerning av uracil i ulike posisjoner i forhold til metyleringen. Forsøkene ble gjennomført på DNA-oligonukleotider med eller uten 5-mC i tillegg til uracil. 5-mC og uracil ble også lokalisert med definerte avstander i forhold til hverandre i samme eller i hver sin DNA tråd. Oligonukleotidene ble så substrat for renset, rekombinant UNG2. Under enzymatisk utkutting av uracil ble det dannet apyrimidine (AP-seter) hvor det ble indusert trådbrudd ved eksponering mot piperidin. Substrat- og produkt-fragmenter ble separert etter størrelse ved hjelp av polyakrylamid gelelektroforese (PAGE) og kvantifisert ved hjelp av ImageQuant. Vi fant at 5-mC to seter oppstrøms og nedstrøms for uracil påvirket aktiviteten til UNG2 på ssDNA, men effekten var ioneavhengig. 5-mC et sete nedstrøms for uracil virket inhiberende på aktiviteten til UNG2 på dsDNA via indirekte påvirkning på sekundærstrukturen. Metylering i motsatt tråd hadde ingen innvirkning på aktiviteten til UNG2. Påvirkningen av metyleringen varierte avhengig av saltinnhold i løsningsbufferen.
dc.description.abstractUNG2 (uracil DNA glycosylase) is the dominant enzyme that removes uracil in the human genome. Methylated cytosine (5-methylcytosine (5-mC)) makes up about 1% of human DNA, and is thus the most common epigenetic modification. The purpose of the study was to investigate whether 5-mC affects UNG2-mediated detection and removal of uracil in different positions relative to the methylation. The experiments were performed on DNA oligonucleotides with or without 5-mC in addition to uracil. 5-mC and uracil were also localized with defined distances relative to each other in the same or in separate DNA strands. The oligonucleotides were then substrate for recombinant UNG2. During enzymatic excision of uracil apyrimidine sites (AP sites) were formed, and strand breakage was induced upon exposure to piperidine. Substrate and product fragments were separated by size by polyacrylamide gel electrophoresis (PAGE) and quantified by ImageQuant. We found that 5-mC two seats upstream and downstream of uracil affected the activity of UNG2 on ssDNA, but the effect was dependent of the ion concentration. 5-mC one seat downstream of uracil inhibited the activity of UNG2 on dsDNA via indirect influence on the secondary structure. Methylation in the opposite strand had no impact on the activity of UNG2. The effect of the methylation varied depending on salinity in the solution buffer.
