Brystkreft er den vanligste kreftformen, og forekomsten av sykdommen er økende på verdensbasis. Proteinet human epidermal vekstfaktorreseptor 2 (HER2) blir overprodusert i flere av brystkrefttilfellene. Derfor er HER2 et viktig mål for utvikling av legemidler som kan forbedre kreftbehandlinger. Hensikten med denne oppgaven har vært å undersøke kjemien rundt, og syntetisere, N-beskyttede pyrrolo[2,3-d]pyrimidin-4-amin derivater som kan inngå i struktur-aktivitetsstudier om HER2 hemming.
Ulike 4-klor-7H-pyrrolo[2,3-d]pyrimidiner ble brukt som utgangsstoffer, og målforbindel-sene ble fremstilt gjennom tre eller fire syntesetrinn. Nitrogenposisjonen i pyrrolringen ble funksjonalisert ved N-alkyleringsreaksjoner, og videre aminering med 3-klor-4-((3-fluorbensyl)oksy)anilin ga fire nye forbindelser. I tillegg ble 6-jod-derivatet brukt i Suzukireaksjoner for å introdusere to nye aryl-grupper. Totalt har dette arbeidet ført til seks nye pyrrolopyrimidiner som kan inngå i struktur-aktivitetsstudier.
Syntesene var stort sett vellykket. N-Alkyleringsreaksjonene var raske og ga høye utbytter. Syrekatalysert nukleofil aromatisk substitusjon var også effektivt for aminering av de fleste pyrrolopyrimidinene, men aminering av 5-jod-derivater var mislykket både ved syre- og base-katalyse. Disse reaksjonene ga størst utbytte av tilsvarende hydrodehalogenert produkt. Flere eksperimenter ble utført for å finne årsaken til denne dehalogeneringen. Undersøkelsene som ble gjort tyder på at omleiring gjennom åpning av pyrrolringen og en retro elektrofil aromatisk substitusjon er mulige årsaker til dehalogeneringen. Videre arbeid er nødvenig for å kunne forstå denne sidereaksjonen. Breast cancer is considered to be the most commonly diagnosed cancer on a global basis, and its burden on society has been rising over the past decades. The human epidermal growth factor receptor 2 (HER2) is overexpressed in a number of breast cancer cases. This protein is therefore an important drug target for the development of treatments. The aim of this thesis has been to investigate the chemistry of, and synthesise, N-protected pyrrolo[2,3-d]pyrimidine-4-amine derivatives for the use in structure-activity relationship (SAR) studies towards HER2 inhibition.
The target compounds were synthesised in three or four steps with 4-chloro-7H-pyrrolo[2,3-d]pyrimidines as starting materials. Functionalization of the pyrrole nitrogen was achieved by N-alkylation, and subsequent amination with 3-chloro-4-((3-fluorobenzyl) oxy)aniline gave four new compounds. Additionally, the 6-iododerivative was arylated by Suzuki cross-coupling to give two new target molecules. Overall, the work provides six new pyrrolopyrimidines for SAR studies towards HER2.
The syntheses towards the target compounds were mostly successful. N-Alkylation proceeded fast and resulted in good yields for all the substrates. Acid catalysed nucleophilic aromatic substitution was efficient for amination of most of the pyrrolopyrimidines, but amination of 5-iododerivatives was unsuccessful both with acid and base as catalysts. Amination of 5-iododerivatives gave the corresponding hydrodehalogenated product as the main compound, and several experiments were performed to investigate the cause of the dehalogenation. The studies performed suggest a rearrangement involving a ring opening of the pyrrole moiety and a retro electrophilic aromatic substitution as possible causes of the dehalogenation. Further work is needed to understand this side reaction.
