Exploring the cGAS-STING immune pathway in infection with respiratory syncytial virus

Respiratorisk syncytialvirus (RSV) er en av hovedårsakene til akutt luftveisinfeksjon. Den sykliske GMP-AMP-syntase (cGAS) - stimulator av interferon-gener (STING)-signalveien gjenkjenner DNA-virus for å stimulere interferon (IFN) antiviral responser, men rollen til cGAS-STING i forhold til RNA virus er lite kjent. Derfor ønsket vi å undersøke den mulige involveringen av cGAS-STING-aksen i in vitro infeksjon med RNA-viruset RSV. Den humane lungefibroblastcellelinjen WI-38 ble valgt som modellsystem siden cGAS-STING regulerer lungebetennelse og fibrotiske mekanismer i menneske- og musemodeller, og RSV-mediert inflammatorisk respons i humane fibroblaster er ikke rapportert.
Våre funn viser at RSV kan replikere og indusere type I IFN-respons i WI-38 fibroblaster. Videre fant vi at RSV-infeksjon førte til økte nivåer av cGAS og STING, sammen med økte nivåer av fosforylert STING, TBK1 og IRF3. Disse resultatene kan tyde på at cGAS-STING-signalveien modulerer RSV-medierte replikasjon og infeksjon. Nedregulering av cGAS ved hjelp av siRNA-mediert tilnærming eller kjemisk hemming, reduserte fosforylert STING og TBK1, noe som antyder at RSV-indusert STING-fosforylering er mediert av cGAS. Interessant nok førte nedregulering av cGAS eller STING til økt RSV-replikasjon i de RSV-permissive cellene HEp-2, mens dette ikke ble observert i WI-38-celler. Tumornekrosefaktor α (TNFα) ble nylig oppdaget å mediere cGAS-STING-aktivering og induksjon av IFN-responser. I samsvar med dette viste vi at rekombinant TNFα aktiverer cGAS-STING-signalveien og øker uttrykket av IFNβ- og IP-10-mRNA. Videre induserte RSV-infeksjon TNFα-mRNA, og økte nivåer av sekretet TNFα-protein. Imidlertid hadde blokkering av TNFα-mediert signalering ved hjelp av infliximab ingen effekt på RSV-stimulert fosforylering av STING, TBK1 eller IRF3. Derfor kan vi ikke konkludere med at TNFα medierer aktivering av cGAS-STING-signalveien i RSV-infiserte celler.
Oppsummert gir resultatene preliminær innsikt i rollen til cGAS-STING-aksen i forhold til RNA-viruset RSV, og viser at TNFα kan aktivere IFN-signalveien via cGAS-STING i RSV- permissive celler. Respiratory syncytial virus (RSV) is a major cause of acute respiratory tract infections. The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway is known to recognise DNA viruses to stimulate interferon (IFN) antiviral responses, but the role of cGAS-STING in relation to RNA viruses is not well understood. In this master`s thesis the potential involvement of the cGAS-STING axis in in vitro infection with the RNA virus RSV was examined. The human lung fibroblast cell line WI-38 was chosen as model system since cGAS-STING regulates lung inflammation and fibrotic mechanisms in human and mice models, and RSV-mediated inflammatory responses in human fibroblasts have not been reported.
We found that RSV is able to replicate and induce type I IFN responses in WI-38 fibroblasts. Moreover, we found that RSV infection led to increased levels of cGAS and STING along with increased levels of phosphorylated STING, TBK1, and IRF3. These results may suggest that the cGAS-STING pathway modulates RSV-mediated replication and infection. Knockdown (KD) of cGAS using siRNA-mediated approaches or chemical inhibition reduced phosphorylated STING and TBK1, suggesting that RSV-induced STING phosphorylation is mediated by cGAS. Interestingly, KD of cGAS or STING led to increased RSV replication in the RSV-permissive cells human HEp-2, whereas this was not observed in WI-38 cells. Tumour necrosis factor α (TNFα) was recently found to mediate cGAS-STING-activation and induction of IFN-responses. Consistent with this, we demonstrated that recombinant TNFα activated the cGAS-STING pathway and increased the expression of IFNβ and IP-10 mRNA. Moreover, RSV infection induced TNFα mRNA and levels of secreted TNFα protein. However, blocking TNFα-mediated signalling using infliximab did not have any effect on RSV-stimulated phosphorylation of STING, TBK1, or IRF3. Hence, we cannot conclude that TNFα mediates activation of the cGAS-STING pathway in RSV-infected cells.
In conclusion, our findings provide preliminary insight into the role of the cGAS-STING axis in relation to the RNA virus RSV and demonstrate that TNFα can activate the IFN-pathway via cGAS-STING in RSV-susceptible cells.
