dc.description.abstract | Rekombinant proteinproduksjon er en stor industri globalt. E. coli er det vanligste
vertsorganismen som brukes til produksjon av rekombinante proteiner.
Oksygenbegrensning i bioreaktorer under produksjon av rekombinante proteiner er en
utfordring som møtes i industrien. Denne masteroppgaven hadde som mål å undersøke
vektbegrensning og metabolsk belastning på E. coli under oksygenbegrensning. En
rekombinert E. coli-stamme, BL21 A2-mCherry, ble brukt i denne masteroppgaven i
tillegg til en villtype stamme, basert på erfaringene fra tidligere arbeid utført av
forskningsgruppen MetaboProt. A2-mCh-stammen inneholdt RK2-plasmidet med
XylS/Pm-promoter systemet, med ampicillin som seleksjonsmarkør og mCherry som
rapporterende protein.
Batchdyrking ble utført i topp-benk bioreaktorer, og prøver ble ekstrahert og analysert
videre. HPLC ble brukt for kvantifisering av organiske syrer og andre ekstracellulære
produkter og substrater, inkludert glukose og etanol. HILIC-MS ble utført for å
kvantifisere intracellulære metabolitter, inkludert ni forskjellige pyridinnukleotider. SDSPAGE
og Western Blot-analyse ble også utført for å kvantifisere produksjonen av
mCherry-protein under dyrkingene.
Resultatene fra denne avhandlingen viser at cellevekst ble redusert av
oksygenbegrensning i bioreaktor-dyrkningene. Imidlertid ble det ikke funnet noen
negativ effekt på mengden proteinproduksjon. I tillegg ble det funnet at glukoseforbruket
var høyere og den observerte biomasseutbyttet lavere for dyrkingene av den
rekombinerte E. coli-stammen med oksygenbegrensning. Det ble funnet svært forhøyede
nivåer av acetat, laktat, format og etanol når villtypen ble utsatt for oksygenbegrensning.
A2-mCh-stammen viste redusert produksjon av laktat og økt produksjon av acetat og
suksinat under oksygenbegrensede dyrkinger. De kvantifiserte dataene for
pyridinnukleotider indikerte en endring i NAD-metabolismen til E. coli-stammene under
oksygenbegrensede dyrkinger, der nivåene av NADPH og NADP ble sterkt redusert ved
senere tidspunkter. Sammenhengen mellom overskuddsmetabolitter og
oksidasjonstilstanden til pyridinnukleotidene blir diskutert med hensyn til skift i
metabolske veier på grunn av anaerobe forhold. Denne masteroppgaven konkluderte
med at problemer med oksygenbegrensning som oppstår i industrien for produksjon av
rekombinante proteiner påvirker metabolske veier og redoksbalansen i E. coli. | |
dc.description.abstract | Recombinant protein production is a large industry worldwide. E. coli is the most
commonly used host organism for recombinant protein production. Oxygen limitation in
the bioreactors during the production of recombinant proteins is a challenge met in the
industry. The present master thesis aimed to investigate growth limitation and the
metabolic burden on E. coli under oxygen limitation. One recombinant E. coli strain, BL21
A2-mCherry, was used in this master thesis in addition to a wild-type strain, based on
the experience of previous work of the MetaboProt research group. The A2-mCh strain
contained the RK2 plasmid with the XylS/Pm promoter system, with ampicillin as the
selection marker, and mCherry as the reporter protein.
Batch cultivations were performed in top-bench bioreactors and samples were extracted
and further analysed. HPLC was used for quantification of organic acids and other
extracellular products and substrates including glucose and ethanol. HILIC-MS was
performed to quantify intracellular metabolites including nine different pyridine
nucleotides. SDS-PAGE and Western Blot analytics was also performed to quantify the
mCherry protein product produced during the cultivations.
The results from this thesis show that cell growth was reduced by oxygen limitation in
the bioreactor cultivations. However, no negative effect on the quantity of protein
production was found. In addition, glucose consumption was found to be higher and
observed biomass yield lower for the oxygen limited cultivations of the recombinant. E.
coli strain. Elevated levels of acetate, lactate, formate and ethanol were found when
exposing the WT strain to oxygen limitation. The A2-mCh strain showed reduced
production of lactate and increased production of acetate and succinate during oxygenlimited
cultivations. The quantified pyridine nucleotide data indicated a shift in the NAD
metabolism of the E. coli strains during oxygen-limited cultivations, where the NADPH
and NADP levels were greatly reduced at later time points. The relation between overflow
metabolites and the redox state of the pyridine nucleotides are discussed in relevance to
shift in metabolic pathways due to the anaerobic conditions. The present master thesis
concluded that issues with oxygen limitation met in the recombinant protein production
industry affect the metabolic pathways and redox balance in E. coli. | |
