Sammenligne differensialtelling av leukocytter i kapillære og venøse prøver på HemoCue WBC DIFF, samt utføre en metodesammenligning av differensialtelling i venøse prøver på HemoCue WBC DIFF og ABX Pentra XL 80

Abstract

Det ble gjennomført en sammenligning av differensialtelling (DIFF) av leukocytter (WBC) i venøst og kapillært blod på HemoCue WBC DIFF, samt en metodesammenligning av DIFF i venøse prøver på HemoCue WBC DIFF og ABX Pentra XL 80. Hensikten var å kartlegge om det er overenstemmelse mellom resultatene av DIFF i venøst og kapillært blod. I tillegg var det ønskelig å undersøke om det er samsvar mellom DIFF på HemoCue WBC DIFF og NTNU Ålesund sitt gjeldende hematologiinstrument, ABX Pentra XL 80.

Til prosjektet ble det samlet inn og utført DIFF på prøver fra 51 deltakere i laboratoriet ved NTNU Ålesund. Av samtlige deltakere ble det tatt én kapillær prøve direkte i mikrokyvette og én venøs prøve i EDTA-rør (etylendiamintetraacetat). For å standardisere forsøket og minimere preanalytiske feil, ble det utarbeidet prosedyrer for prøvetaking og analyse på HemoCue WBC DIFF.

Ved sammenligningen av DIFF i kapillære og venøse prøver på HemoCue WBC DIFF var det god overensstemmelse, men dette gjaldt kun for analyttene total WBC og nøytrofile granulocytter. HemoCue WBC DIFF kan derfor ikke benyttes for en komplett fem-parts WBC DIFF, men heller som kontroll av total WBC og nøytrofile granulocytter. Ved metodesammenligning fremkom det god sammenheng mellom de to instrumentene når det gjaldt de to samme analyttene, total WBC og nøytrofile granulocytter.

A comparison of the differential count (DIFF) of leukocytes (WBC) in venous and capillary blood was carried out on HemoCue WBC DIFF, as well as a method comparison of DIFF in venous samples on HemoCue WBC DIFF and ABX Pentra XL 80. The purpose was to map whether there is an agreement between the results of DIFF in venous and capillary blood. In addition, it was desirable to investigate if there was agreement between DIFF on HemoCue WBC DIFF and NTNU Ålesund current hematology instrument, the ABX Pentra XL 80.

For the project, samples from 51 participants were collected and analyzed in the laboratory at NTNU Ålesund. From all participants, one capillary sample was directly drawn into a microcuvette and one venous sample in an EDTA tube (ethylenediaminetetraacetic acid). In order to standardize the experiment and minimize preanalytical errors, procedures for sampling and analysis on the HemoCue WBC DIFF were created.

When comparing DIFF in capillary and venous samples on the HemoCue WBC DIFF, there was good agreement. However, this only applied to the analytes total WBC and neutrophil granulocytes. Therefore, we concluded that the HemoCue WBC DIFF cannot be used for a complete five-part WBC DIFF, but rather as a control for total WBC and neutrophil granulocytes. When comparing the methods, there was a good correlation between the two instruments when it came to the same two analytes, total WBC and neutrophil granulocytes.
