The effect on quality and shelf life of Atlantic salmon treated with natural antioxidants combined with modified atmosphere packaging

Sammendrag
Etterspørselen etter naturlige og ferske fiskeprodukter av høy kvalitet er økende. Atlanterhavslaks tilfører flere viktige næringsstoffer til kostholdet, men bruken er begrenset grunnet høy sensitivitet til biokjemisk og mikrobiell bedervelse. Tidligere studier har vist at modifisert atmosfærepakking (MAP) og tilsetning av antioksidanter kan forsinke mikrobiell vekst og hemme lipidoksidering. Målet med denne studien var å undersøke kombinasjonen av de to pakkemetodene for å bevare kvalitet og forsinke lipidoksidering i fisk.
Antioksidantløsninger fra naturlige kilder (oregano og dill) ble tillaget før antioksidantaktivitet og fenolinnhold ble analysert ved bruk av metodene DPPH, ABTS og Folin-Ciocalteu. Atlanterhavlaks pakket med vakuum, MAP (CO2:N2 60:40) og MAP i kombinasjon med de tillagde antioksidantløsningene ble lagret kaldt (4 ± 1˚C) i 0-16 dager. Endringer i kvalitet under lagring ble analysert i form av drypptap, mikrobiell vekst, farge, pH og proteinløselighet. Primære og sekundære oksidasjonsprodukter ble målt ved PV, CD og TBARS. I tillegg ble flyktige forbindelser analysert ved bruk av GC-MS, SIFT-MS og SESI-MS.
Vakuumpakkede prøver hadde høyere drypptap, høyere tap av saltløselige proteiner og lavere pH. Ingen signifikante forskjeller i mikrobiell vekst ble funnet, og grenseverdien for inntak ble overskredet etter 6 og 8 lagringsdager. Oreganoløsningen var forventet å være den mest framtredende antioksidanten. Prøver pakket med MAP + dill hadde lavere verdier av PV og TBARS, men ingen konklusjon ble trukket grunnet mangel på signifikante forskjeller. CD-indeksen viste seg ikke å være gunstig til overvåkning av oksidasjonsstatus over tid. Når det kommer til kvalitetsparametere førte tilsats av oregano til lysere farge i prøvene, mens dill førte til høyere pH og mindre tap av saltløselige proteiner. 29 flyktige organiske forbindelser ble detektert i luftrommet til prøvene som kom fra både lipidoksidering og mikrobiell aktivitet. Fem forbindelser ble foreslått som mulige markører for bedervelse når produksjonen i laks er forstått.
Til oppsummering viste denne studien fordeler ved bruk av MAP sammenlignet med vakuum når det gjelder bevaring av kvalitet i laks. Antioksidanter kombinert med MAP påvirket også kvalitetsparameterene i noen grad. Etter bestemmelse av antioksidantaktivitet ble det antydet at oregano hadde større potensial til å forsinke lipidoksidering i maten sammenlignet med dill, men dette studiet klarte ikke å oppdage store forskjeller mellom antioksidantløsningene i praksis. Consumers show an increased demand for natural, fresh and high quality fish products. Atlantic salmon provides several important nutrients to the human diet including essential n-3 fatty acids, but the use is limited due to its high susceptibility to biochemical and microbial spoilage. Earlier studies have shown that modified atmosphere packaging (MAP) and antioxidant addition can delay microbial growth and retard lipid oxidation. The aim of the current study was to investigate the combination of natural antioxidants and MAP in order to retain the quality and delay lipid oxidation in fish.
Antioxidant solutions obtained from natural sources (oregano and dill) were prepared and examined for antioxidant activity and phenolic compounds using the DPPH, ABTS and Folin-Ciocalteu assays. Atlantic salmon treated with vacuum, MAP (CO2:N2 60:40) and MAP in combination with the prepared antioxidant solutions was stored at chilled conditions (4 ± 1˚C) for 0-16 days. Quality changes during storage were analysed in terms of drip loss, microbial growth, colour, pH and protein solubility. Primary and secondary oxidation products were measured by PV, CD and TBARS. In addition, volatile compounds were analysed using GC-MS, SIFT-MS and SESI-MS.
Vacuum treated samples had higher drip loss values, higher loss of salt soluble proteins and lower pH. No significant differences in microbial growth were found, and the consumption limit was reached after 6 and 8 days of storage. The oregano solution was predicted to possess the highest antioxidant properties. Samples treated with MAP + dill showed lower values of PV and TBARS, but no conclusions were drawn due to lack of significant differences. The CD index did not prove to be effective in showing oxidation status over time. Considering quality parameters, oregano treatment led to lighter colour of the samples, while dill treatment led to higher pH and lower loss of salt soluble proteins. 29 VOCs were detected in the headspace of the samples originating from both lipid oxidation and microbial activity. Five compounds were proposed as possible markers of spoilage when their production in salmon is understood.
To conclude, this study showed advantages of using MAP compared to vacuum in terms of retaining quality of salmon. Antioxidants combined with MAP also influenced the parameters to some extent. Antioxidant activity determination suggested that oregano had higher potential to cause a delay in lipid oxidation in the food system compared to dill, but the study was unable to detect large differences between the antioxidant solutions in practice.
