For å motvirke abiotisk stress, akkumulerer mikroorganismer osmobeskyttendemolekyler, også kalt osmolytter, ved enten de novo biosyntese eller opptakfra omgivelsene. Den intracellulære akkumuleringen av disse molekylenemed lav molekylærvekt har vist seg å øke motstanden mot osmotisk stress,temperaturendringer, oksygenradikaler og stråling, i tillegg til å stabiliseretertiærstrukturen til intracellulære makromolekyler som proteiner eller dobbelttrådetDNA. Di-myo-inositol-fosfat (DIP) er en osmolytt som mistenkes å halovende egenskaper som UV-beskyttelse, fuktigivning, PCR-forsterkelse og DNAbeskyttelse.Per i dag produseres osmolytter kun industrielt vedbruk av ekstremofile mikroorganismer via ineffektive prosesser. Ulempene medslike prosesser inkluderer forkortet levetid på utstyr, redusert volumetrisk utbytteog kompliserte opp- og nedstrømsprosesser. Utvikling av fermenteringsprosessermed lav salinitet er ønskelig, der heterolog ekspresjon i mesofile organismer villetillate slike forbedringer. Produksjonen av DIP er ennå ikke etablert i mesofilemikroorganismer, og ettersom studier av ekstremolytter krever spesialisert utstyr,vil etablering av et mesofilt produksjonssystem forenkle produksjonen av DIP pålaboratorie-skala. Corynebacterium glutamicum er en veletablert mikrobiell arbeidshest,mest kjent for storskala produksjon av aminosyrer. Bakterien har alleredeblitt genetisk modifisert for produksjon av osmolytter som ektoin, hydroksyektoin,mannosylglycerat, og L-pipekolsyre. I dette arbeidet har en C. glutamicumvilltype blitt genetisk modifisert for å uttrykke DIP-biosynteseveien. Dette bleoppnådd ved å kombinere gener fra hypertermofilene Thermotoga maritima ogPyrococcus furiosus. For å teste de nykonstruerte stammene ble fermenteringer imikrobioreaktorer utført under osmotisk og termisk stress. Ved dyrking av stammenei minimalt medium supplert med 0,5 M NaCl eller 1 M NaCl ved 30◦C, er detkun stammen som bærer det DIP-biosyntetiske operonet fra T. maritima som vistevekstforbedringer med hensyn til veksthastighet og biomassedannelse. Deretter ble enristekolbe-fermentering, kombinert med osmotisk-sjokkfall, utført for ekstraksjon avflere osmolytter inkludert DIP. Til sist ble stammen med best ytelse testet i bioreaktorerpå laboratorieskala med samme osmotiske sjokkfall tilnærming. Forsøket beviste atden anvendte bioprosessen var funksjonell for produksjon av ikke-native osmolytter. To combat abiotic stresses, microorganisms accumulate compatible solutes by either denovo biosynthesis or uptake from the environment. The intracellular accumulationof these low-molecular weight compounds have been found to increase resistanceto osmotic stress, temperature change, oxygen radicals, and radiation, in additionto stabilize the tertiary structure of intracellular macromolecules such as proteinsor double-stranded DNA. Di-myo-inositol-phosphate (DIP) is a compatiblesolute suspected to possess promising features as a UV protectant, moisturizer,PCR enhancer, and DNA protectant. As of today, compatiblesolutes are only industrially produced using extremophile microorganisms followinginefficient processes. The drawbacks to such processes include shortened lifespanof equipment, reduced volumetric yield, and complicated up and downstreamprocesses. Development of low-salinity fermentation processes is desired, whereheterologous expression in mesophile organisms would allow such improvements.The production of DIP has yet to be established in a mesophile system. As the studyof extremolytes requires specialized equipment, the establishment of a mesophilesystem would simplify the production of DIP at the lab-bench scale. Corynebacteriumglutamicum is a well-established microbial workhorse, best known for its large-scaleproduction of amino acids. It has already been genetically engineered for theproduction of compatible solutes such as ectoine, hydroxyectoine, mannosylglycerate,and L-pipecolic acid. In this work, C. glutamicum has been geneticallyengineered for the heterologous expression of the DIP biosynthesis pathway. Thiswas achieved by combining genes from the hyperthermophiles Thermotoga maritimaand Pyrococcus furiosus. To test the newly constructed strains, fermentations inmicrobioreactors were performed under osmotic and thermal stress. When cultivatingthe strains in a minimal medium supplemented with 0.5 M or 1.0 M NaCl at 30◦C,only the strain carrying the DIP biosynthetic operon from T. maritima showed growthimprovements with regards to growth rate and biomass formation. Next, a shakeflask fermentation, coupled with an osmotic shock down approach was performed forthe extraction of compatible solutes including DIP. Finally, the best performing strainwas tested in lab-scale bioreactors following the same osmotic shock down approach,proving the feasibility of this bioprocess for the production of non-native compatible solutes.
