Photosensitizing effects of Al(III) Phthalocyanine Chloride Disulfonic Acid (AlPcS2a) – in vitro studies on AY-27 and T-24 cells

Fotodynamisk terapi (PDT) er en behandlingsmetode som benytter et fotosensibiliserende stoff (PS) som induserer celledød ved aktivering av lys. Fotokjemisk internalisering (PCI) er en ny teknologi som bygger videre på de samme biofysiske prinsippene som PDT, men hvor hovedhensikten er å forbedre leveringen av for eksempel kjemoterapeutiske midler til cytosol i cellene. Blærekreft behandles i dag vanligvis med kirurgisk fjerning eller adjuvant terapi, men har potensialet til å behandles med PDT og PCI. Behandlingsmetodene som benyttes i dag fører ofte til bivirkninger som kan reduseres dersom PDT og PCI benyttes i stedet. I dette prosjektet ble lokalisasjonen og den fotosensitive effekten av den kommersielle PSen, Al(III) phthalocyanine chloride disulfonic acid (AlPcS2a), studert i henhold til blærekreftcellelinjene AY-27 og T-24 fra henholdsvis rotte og menneske. Cellene ble eksponert for AlPcS2a (1 µM) og belyst med ulike lysdoser av rødt lys (633 nm, 21mW/cm2 - 36mW/cm2). Evnen PSen har til å svekke levedyktigheten til cellene ble evaluert ved hjelp av AlamarBlue-analysemetoden. Et konfokalmikroskop ble benyttet for å observere lokalisasjonen av AlPcS2a i AY-27 og T-24 cellene, og arealoverlapp mellom PSeren og forskjellige fluoriserende fargestoff ble beregnet.
Resultatene viste at AlPcS2a ble tatt opp i cellene, og at PSen primært lokaliserte seg i lysosomer i begge cellelinjene, med et arealoverlapp på 42.8% +- 3.6% og 30.3% +- 2.8% i henholdsvis AY-27 og T-24 cellene. PDT-eksperimentene viste at AlPcS2a var en effektiv PS for begge cellelinjene, som førte til økt celledød under økende eksponering av rødt lys. Median dødelig dose (LD50) ble funnet rundt tre minutter for AY-27 cellene, og alle cellene var døde etter ni minutter med belysning. For T-24 cellene ble LD50 funnet rundt seks minutter, og etter 15 minutter var over 80% av cellene døde. Fra disse resultatene kan det konkluderes med at AlPcS2a er effektiv for å indusere AY-27 og T-24 celledød, og dermed er en potensiell PS for PDT for blærekreft. Videre studier er nødvendig for å fastlegge PCI-teknologien ved å bruke AlPcS2a i en rotteblærekreftmodel, men at AlPcS2a lokaliseres i lysosomer er lovende. Photodynamic therapy (PDT) is a treatment that utilizes a photosensitizing molecule (PS) that induces cell death upon light activation. Photochemical internalization (PCI) is a novel technology building on the same physical principles as PDT, but where the aim is to improve the release of e.g. therapeutic molecules from endocytic vesicles into the cytosol of the cells. Bladder cancer is usually treated with surgical removal and adjuvant therapy, but has the potential to be treated with PDT and PCI. Today's treatment methods often induce several side effects that can be reduced by using PDT and PCI instead. In this work, localization and photosensitizing effects of the commercial PS, Al(III) phthalocyanine chloride disulfonic acid (AlPcS2a), were explored in relation to the bladder cancer cell lines AY-27 and T-24 from rat and human, respectively. The cells were exposed to AlPcS2a (1 µM) and given different light doses using a red light source (633 nm, 21mW/cm2 - 36mW/cm2). The capacity of the PS to impair viability of the cells was evaluated by means of the AlamarBlue assay. A laser scanning confocal microscope was used to observe the localization of AlPcS2a in AY-27 and T-24 cells, and the area overlap between the PS and different fluorescent dyes was calculated.
The results showed that the cell lines exhibited uptake of AlPcS2a and that the PS dominantly localized in lysosomes in both cell lines, with an area overlap of 42.8% +- 3.6% and 30.3% +- 2.8% for AY-27 and T-24 cells, respectively. The PDT experiments demonstrated that AlPcS2a was effective against bladder cancer cells, with an increased cell death during increased red light exposure. The median lethal dose (LD50) was found to be around three minutes of light exposure for AY-27 cells, and all cells were killed after nine minutes. For the T-24 cells, LD50 was found to be around six minutes, and after 15 minutes more than 80% of the cells were dead. From these results, it was concluded that AlPcS2a is effective in causing damage to AY-27 and T-24 cancer cells, and is a potential PS for PDT of bladder cancer. Further studies are necessary to establish the PCI technology by using AlPcS2a in a rat bladder cancer model, but the findings that AlPcS2a primarily recruited to lysosomes are promising results.
