dc.description.abstract | Formålet med dette masterprosjektet var å utforske fotodynamiske, fotofysiske og fotokjemiske
egenskaper til noen nye glykosylerte kloriner. Dette prosjektet er en del av et
pågående samarbeid mellom Institutt for fysikk, NTNU, og Institutt for kjemi, Linköpings
universitet, Sverige. Samarbeidet tar sikte på å undersøke egenskapene til syv nye glykosylerte
kloriner (KA20-familien), syntetisert av Dr. Katriann Arja i Prof. Peter Nilssons
gruppe, Linköpings universitet. Dette masterprosjektet fokuserte på følgende to kloriner:
KA20-11H-proto og KA20-11H forbundet med glukose, de ble undersøkt for sine fotofysiske,
fotokjemiske og fotodynamiske egenskaper i to kreftblærecellelinjer, AY-27 (rotte) og T-24
(menneske).
Optisk spektroskopi ble brukt for å finne absorpsjons- og emisjonsspektrene til klorinene
og deres respektive kvanteutbytte (QY) og singlett oksygen-kvanteutbytte i forskjellige
løsningsmidler. Begge klorinene hadde absorpsjon i den blå delen (Soret-båndet) og den
røde delen(Q-båndet) av spekteret. KA20-11H-proto (0,14 +\- 0,01) ble funnet å ha en
høyere QY enn KA20-11H (0,045 +\- 0,004). De hadde en lignende singlett oksygen-kvanteutbytte på 0,36 (KA20-11Hproto)
og 0,38 (KA20-11H) i kloroform.
Cytotoksisitetsanalyser ved bruk av AlamarBlue ble utført på begge cellelinjene med
klorinene. Overlevelseseksperimenter ved bruk av AlamarBlue post fotodynamisk terapi
(PDT) med rødt lys (633 nm) ble utført på AY-27 og T-24 celler med 5 uM og 10 uM løsninger.
Flowcytometri ble også utført for å undersøke celledød etter PDT. Flowcytometerforsøk ble
også utført etter cytotoksisitetseksperiment på T-24 celler etter inkubasjon med KA20-11H.
Konfokalbilder ble tatt av suspensjonene som ble brukt til flowcytometeranalyse for å
bekrefte opptak av fargestoffene. Konfokalbilder ble også tatt av begge cellelinjer inkubert
med KA20-11H (15 uM) og KA20-11H-proto (5 uM) for å bekrefte opptak og for å
se etter visuelle tegn på belysning. Prosjektet viste fotodynamiske potensial til de nye
glykosylerte klorinene ved bruk av rødt lys, ettersom de effektivt induserte celledød i både
AY-27 og T-24 cellelinjene, og hadde en lav mørk toksisitet i begge cellelinjene. | |
dc.description.abstract | The aims of this master project were to explore photodynamic, photophysical and photochemical
properties of some novel glycosylated chlorins. This project is a part of an
ongoing collaboration between Department of Physics, NTNU, and Department of Chemistry,
Linköping University, Sweden. This collaboration aims to examine properties of seven
novel glycosylated chlorins (KA20 family) synthesised by Dr. Katriann Arja in Prof. Peter
Nilsson’s group, Linköping University. This master project focused on the following two
chlorins: KA20-11H-proto and KA20-11H complexed with glucose, they were examined for
their photophysical, photochemical and photodynamic properties in two cancer bladder
cell lines in this project, AY-27 (rat) and T-24 (human).
Optical spectroscopy was used to find the absorption and emission spectra of the chlorins
and their respective quantum yield (QY) and singlet oxygen quantum yield in different
solvents. Both chlorins were found to have absorption in the blue (Soret band) and red
(Q band) regime and had a similar singlet oxygen QY of 0.36 and 0.38 in chloroform. KA20-11H-proto
(0.14 +\- 0.01) was found to have a higher QY than KA20-11H (0.045 +\- 0.004).
Cytotoxicity assays using AlamarBlue were performed on both cell lines with chlorins.
Survival experiments using AlamarBlue post Photodynamic therapy (PDT) with red light
(633nm) were performed on AY-27 and T-24 cells using 5 uM and 10 uM solutions. Flow
cytometry was also performed to examine cell death pathways after PDT incubated with
photosensitizer (PS) (5 uM). The flow cytometer experiment was also performed after
a cytotoxicity experiment on the T-24 cell line using KA20-11H. Confocal laser scanning
microscope (CLSM) images were taken of the suspensions used for flow cytometer analysis
to confirm uptake of the dyes. The CLSM studies were also performed for both cell lines
incubated with KA20-11H (15 uM) and KA20-11H-proto (5 uM) to confirm uptake and
look for visual signs after illumination.
The project proved the photodynamic potential of the novel PSs using red light as they
were effective inducing cell death in both the AY-27 and T-24 cell lines and a low dark
toxicity was observed in both cell lines. | |
