dc.description.abstractBiofilmproduksjon er en viktig fenotypisk egenskap hos en rekke bakteriearter sett i sammenheng med behandling av infeksjoner. I denne oppgaven ble det etablert og validert en semikvantitativ metode for fenotypisk påvisning av biofilmproduksjon i S. epidermidis og S. aureus basert på eksisterende litteratur av Christensen et al., Massey et al. og Ziebuhr et al. Metoden ble validert med hensyn på inkubasjonstid, vaskeprosedyre, fiksering, fargetid og spektrofotometrisk deteksjon av biofilmmengde. Formålet med undersøkelsene var å finne alternativene som var mest praktisk gjennomførbar, sparte mest tid og opprettholdt god analytisk kvalitet. Resultatet indikerer at 18-24 timer inkubasjon, 3 vaskerunder med springvann i automatisert platevasker, 15 minutters fikseringstid med 96% etanol, 15 minutters fargetid med 0,1% krystallfiolett, 7% eddiksyre som suspensjonsmiddel og avlesningsbølgelengde på 590 nm er egnet til semikvantitativ påvisning av biofilmproduksjon hos S. aureus og S. epidermidis. Presisjonen på de tre kontrollstammene lå henholdsvis på 27,2%, 21,5% og 23,3%. Verdiene sett i sammenheng med en semikvantitativ metode er ikke dårlige. Resultatene viser at metoden tydelig skiller mellom ingen, moderat og høy biofilmproduksjon hos kontrollstammene av S. epidermidis. Bestemmelse av biofilmproduksjonen hos invasive versus ikke-invasive isolater viser at ingen av bakteriestammene har høy tendens til biofilmproduksjon. Variasjonen innad de invasive stammene ligger på 72,0% sett i sammenligning med 37,3% variasjon innad de ikke-invasive. Dette indikerer at enkelte av bakteriestammene kan ha en voldsom produksjon i forhold til resten av den analyserte populasjonen. 23,3% av de invasive stammene hadde et nivå av biofilmproduksjon over negativ kontroll, i forhold til kun 6,3% av ikke-invasive stammer. Undersøkelsen i har vist i sin helhet at metoden kan benyttes som mål på biofilm produksjon i S. epidermidis og S. aureus. Videre arbeid er å sjekke reproduserbarheten på kliniske bakteriestammer over en lengre tidsperiode.
dc.description.abstractBiofilm production is an important phenotypic property in several bacterial species, associated with treatment of infections. In this thesis, a semi-quantitative method for phenotypic detection of biofilm production in S. epidermidis and S. aureus was established and validated based on existing literature by Christensen et al., Massey et al. and Ziebuhr et al. The method was validated with consideration to incubation length, washing procedure, fixation, staining duration, and spectrophotometric detection of biofilm product. The purpose of which was to find the alternatives that were the most practically feasible, least time consuming and maintained a good level of analytical quality. The results indicate that 18-24 hours of incubation, 3 rounds of automated washing with the use of tap water, 15 minutes of fixation with 96% ethanol, 15 minutes of staining with 0.1% crystal violet, 7% acetic acid as suspending agent and reading wavelength of 590 nm are suitable for semi-quantitative detection of biofilm production in S. aureus and S. epidermidis. The precision measurement of the three control strains; non-, moderate- and high biofilm producing, were 27.2%, 21.5% and 23.3%, respectively. The values, when seen in the context of a semi-quantitative method were not inadequate. The results show that the method clearly distinguishes between non, moderate, and high biofilm production in the control strains of S. epidermidis. Determination of biofilm production in invasive versus non-invasive isolates of S. aureus shows that none of the bacterial strains have a high tendency for biofilm production. The variation within the invasive strains is 72.0% compared to 37.3% variation within the non-invasive strains. This is an indicator that some of the invasive bacterial strains can have an extreme production level in comparison to the rest of the sample population. It was also determined that 23.3% of the invasive strains had a level of biofilm production which was over the negative control, compared to only 6.3% of non-invasive strains. In its entirety this study has shown that the method can be used to measure biofilm production levels in S. epidermidis and S. aureus. Further work, to validate the reproducibility of clinical bacterial strains over a longer period is needed.
