dc.description.abstract | For å studere endring i proteinekspresjonen av ulike SIGLEC proteiner, ble A549 cellelinjen
brukt. Cellekulturen ble dyrket i DMEM/F-12 + 10% FBS + 1%AA dyrkingsmedium, samt
ulike konsentrasjoner av sialinsyre.
I begynnelsen av forsøket gikk mye av arbeidet ut på å optimalisere prosedyren for
proteinekspresjon av SIGLEC under SDS-PAGE. Celleprøvene som ble benyttet under
forsøkene hadde blitt dyrket på forskjellige konsentrasjoner av sialinsyre, disse var 0 mM, 0,1
mM, 1 mM og 2 mM.
En betydelig del av forsøket gikk ut på å optimalisere metodeutviklingen. Gjennom denne ble
det testet ulike mengder med radioimmunoprecipitation assay (RIPA) buffer, forholdet på
2,5:1 RIPA lyseringsbuffer til prøve ga best resultat. Videre ble det benyttet to ulike SDSPAGE gel konsentrasjoner, 12% og 16%, for å finne gelen som var mest optimal for
proteinuttrykk av SIGLEC. Avgjørelsen falt på 12% SDS-PAGE gelen da denne ga klarere
og bedre visualiserte proteinbånd enn 16% SDS-PAGE gelen.
Resultatene av prøvene dyrket på forskjellige konsentrasjoner av sialinsyre viste lite til ingen
endring i proteinekspresjonsmønster. Det ble konkludert at metoden som ble benyttet ikke ga
tilstrekkelig med informasjon om de ulike proteinene i prøvene, anbefaler videre testing med
western blotting. | |
dc.description.abstract | When studying the changes in the protein expression of different SIGLEC proteins, the cell
line A549 was used. These cells were cultivated in DMEM/F-12 + 10% FBS + 1%AA culture
medium with differing concentrations of sialic acid.
At the beginning of the experiment, a significant amount of the work went into optimizing the
procedure used to visualize protein expression of SIGLEC with SDS-PAGE. The samples
were cultivated at different concentrations of sialic acid, these concentrations were 0 mM, 0,1
mM, 1 mM and 2mM.
A significant portion of the experiment was the optimization of methods. Different volumes
of radioimmunoprecipitation assay (RIPA) buffer were tested and the concentration of 2,5:1
RIPA buffer to test sample was found to give the best results. This was used with different
concentrations of SDS-PAGE gels to find the best one for protein expression of SIGLEC
proteins. 12% and 16% concentrations were tested and 12% SDS-PAGE gel was found to
provide a clearer visualization of the protein bands than the 16% SDS-PAGE gel.
The results of the samples cultivated on different concentrations of sialic acid showed little to
no change in the protein expression of the SIGLEC proteins. These results suggest that not
enough information were given by this method. It is therefor recommended to try a different
method such as western blotting. | |
