Metabolic Profiling of the Glioblastoma Cell Line U-87 MG in Response to Hypoxia and HIF-1alpha Inhibitors

Glioblastom er en krefttype med høy dødelighet, og dens dødelighet har sammenheng med graden av hypoksi. Hypoksi, fraværet av nok oksygenering av vev, vil aktivere en rekke tilpasninger som fører til økning i behandlingsresistens via stabiliseringen av transkripsjonsfaktoren HIF-1α. Målet i dette studiet er å undersøke hvordan hypoksi påvirker den metabolske profilen i glioblastom in vitro ved hjelp av cellelinjen U-87 MG. Videre ble også tilsats av HIF-1α-inhibitorer testet, for å finne ut om de kan motvirke eller endre den hypoksiske responsen.
Først ble viabiliteten i U-87 MG testet med fire ulike HIF-1α-inhibitorer: 17-AAG, Everolimus, chrysin og YC-1 i normoksi og hypoksi. Cellenes viabilitet varierte med inhibitor, men det var liten forskjell mellom normoksiske og hypoksiske celler. Etterpå, ved hjelp av ELISA, ble nivå av HIF-1α målt i hypoksiske og normoksiske celler. Celler i normoksi viser en tilstedeværelse av HIF-1α, men hypoksi induserer en 12-15% økning av HIF-1α etter åtte timer. For å teste om HIF-1α ville gå ned med en inhibitor, ble 17-AAG og Everolimus testet i hypoksi med ELISA. HIF-1α viste en nedgang i cellene på 16% og 28% ved å bruke henholdsvis 17-AAG og Everolimus. Etter inhibitorenes effekt på celleviabilitet og HIF-1α-nivå var funnet, ble den metabolske profilen undersøkt. 1H-NMR ble brukt for å måle glukose- og glutaminforbruk, samt laktatutskillelse. 1H-NMR avklarte lignende rater av glukoseopptak og laktatutskillelse i den hypoksiske og normoksiske kontrollen. Celler behandlet med en av inhibitorene viste en økning i glukoseopptak og laktatutskillelse i hypoksiske forhold. Glutaminopptak gikk generelt ned i hypoksi, og 17-AAG økte glutaminopptaket i normoksi i forhold til kontrollen. Intracellulære metabolitter ble kvantifisert ved hjelp av capIC-MS/MS og LC-MS/MS. Sammenlignet med normoksiske forhold, økte de glykolytiske metabolittene med alle behandlinger i hypoksi. Samtidig sank mellomproduktene i sitronsyresyklusen i nivå, noe som indikerer en hypoksisk glykolytisk respons. 17-AAG-behandlede celler viste en økning i pyrimidinnukleosidfosfater i hypoksi. Ved å undersøke inhibitorenes metabolske profil sammenlignet mot kontrollen i samme oksygenspenning, avtok konsentrasjonen av metabolitter i både glykolysen og sitronsyresyklusen. Selv om hypoksi økte glykolytiske metabolitter totalt sett, forårsaket inhibitorene en reduksjon i glykolytiske mellomprodukt sammenlignet med kontrollen i både normoksi og hypoksi. Onkometabolitten 2-hydroksyglutarat viste også en signifikant nedgang etter inhibitorbehandling. Everolimus viste en nedgang i metabolitter relatert til beskyttelse mot oksidativt stress.
Oppsummert, viser U-87 MG-cellelinjen en pseudohypoksisk fenotype i normoksi i henhold til dens nivå av HIF-1α og like nivå av glukoseopptak og laktatutskillelse sammenlignet med hypoksi. På den andre siden, når cellene er utsatt for hypoksi, øker både HIF-1α og glykolytiske intermediat enda mer. Starten av hypoksi er den største bidragsyteren for den metabolske profilen, men tilsats av inhibitorer gir en særskilt separasjon av metabolittsammensetning i behandlede celler fra de ubehandlede. Begge inhibitorer forårsaker en nedgang i vekst, glykolytiske intermediat og onkometabolitten 2-hydroksyglutarat, og motvirker den hyposiske responsen til en viss grad. Denne studien viser den bemerkelsesverdige metabolske plastisiteten hos celler, og hvordan hypoksi og inhibitorer kan endre den metabolske profilen hos glioblastomceller. Glioblastoma is a cancer with a high mortality rate, and which mortality is correlated with the degree of hypoxia. Hypoxia, the absence of proper oxygenation of tissues, will activate a series of adaptations leading to increased resistance to treatments through the stabilization of the transcription factor HIF1α. The aim of this study is to investigate how hypoxia affects the metabolic profile of glioblastoma in vitro, using the cell line U-87 MG. Furthermore, the addition of inhibitors of HIF-1α was also tested, to elucidate if they can counteract or change the hypoxic response.
Firstly, the viability of U-87 MG was tested with four HIF-1α inhibitors: 17-AAG, Everolimus, chrysin and YC-1 in normoxia and hypoxia. Overall, cells’ viability varied with inhibitor, but there was little difference between normoxia and hypoxia. Thereafter, using ELISA, levels of HIF-1α were measured in hypoxic and normoxic cells. Cells in normoxia show presence of HIF-1α, but hypoxia induces a 12-15% increase of HIF-1α after eight hours. In order to test whether HIF-1α would go down with an inhibitor, 17-AAG and Everolimus were tested in hypoxia with ELISA. HIF-1α proved to decrease in the cells with 16% and 28% using 17-AAG and Everolimus respectively. After having established the inhibitors’ effect on cell viability and HIF-1α, the metabolic profile was studied. 1H-NMR was used to measure glucose and glutamine consumption, and lactate excretion. 1H-NMR uncovered similar rates of consumption and excretion in the hypoxic and normoxic control. Cells treated with either inhibitor showed an increase in glucose consumption and lactate excretion in hypoxic conditions. Glutamine consumption went down in hypoxia, and 17-AAG increased the glutamine consumption in normoxia compared to the control. The intracellular metabolites were quantified using capIC-MS/MS and LC-MS/MS. Relative to normoxic conditions, the glycolytic metabolites increased with all treatments in hypoxia, while the TCA cycle intermediates were lowered, indicating a hypoxic glycolytic response. 17-AAG-treated cells showed an increase in pyrimidine nucleoside phosphates in hypoxia. Studying the metabolic profile of cells treated with inhibitors relative to the control in the same oxygen tension, both the glycolytic metabolites and TCA cycle intermediates went down in concentration. Even though hypoxia increased glycolytic intermediates overall, inhibitors caused a decrease in glycolytic intermediates compared to the control in both normoxia and hypoxia. The oncometabolite 2-hydroxyglutarate also showed a significant decrease after inhibitor treatment. Everolimus showed a decrease in metabolites related to protection against oxidative stress.
Summarized, the U-87 MG cell line displays a pseudohypoxic phenotype in normoxia according to its levels of HIF-1α and equal levels of glucose consumption and lactate excretion compared to hypoxia. On the other hand, when exposed to hypoxia, both HIF-1α and the glycolytic intermediates increase more. The onset of hypoxia is the major determining factor for the metabolic profile, but the addition of inhibitors distinctly separates the metabolite pools in treated cells from the untreated. Both inhibitors reduce levels of growth, glycolysis and the oncometabolite 2-hydroxyglutarate, and counteract the hypoxic response to a certain degree. This study showcases the remarkable metabolic plasticity of cells, and how hypoxia and inhibitors significantly can change the metabolic profile of glioblastoma cells.
