dc.description.abstract | Stressresponser representerer dyrets reaksjon i utfordrende situasjoner og blir brukt som en
velferdsindikator. Kortisol er relevant for fiskevelferd og er en ofte målt komponent fra det
primære stressresponssystemet. Målet for bacheloroppgaven er å se om vi kunne forenkle
Neogen-kittet, ved å erstatte ekstraksjonstrinnet med SDR fra Enzo-kittet. Problemstillingen
for oppgaven er «Kan Neogen-kittet forenkles, og samtidig beholde nøyaktigheten i måling av
kortisol?». Det er viktig å kvalitetssikre at resultatene fra forsøket er repeterbare ved å sjekke
korrelasjonen mellom to plater. Både ekstraherte- og ikke-ekstraherte prøver viste korrelasjon
mellom platene. Deretter optimaliserte vi ekstraheringsmetoden der ekstraheringsvarianten med
to minutt blanding ble utvalgt. Det neste trinnet var en stresstest, hvor en testet de ulike
metodene med samme prøver fra fisk før og under stresspåvirkning. Testen gir en sterk
antydning på forskjell i kortisolnivå mellom før og under stress. Til slutt så en etter korrelasjon
og forskjeller mellom den ekstraherte og ikke-ekstraherte metoden, dette viser om det er mulig
å forenkle Neogen-metoden og samtidig beholde den nøyaktige kortisolmålingen. Resultatet
viste korrelasjon mellom metodene og at prosessen ble forenklet, samt en unngår
ekstrakostnadene for Enzo-kittet. Imidlertid er det også en forskjell mellom metodene siden
null-hypotesen ble forkastet pga. lavere konsentrasjon hos ikke-ekstraheringsmetoden.
Fremtidige studier burde fokusere på å finne en SDR som er mer kompatibel med Neogenkittet. Det anbefales også å bruke flere prøver for å få en tydeligere variasjon mellom gruppene
og unngå at ekstremverdier påvirker resultatet negativt. En burde også finne den optimale
fortynningen som gir resultat under 10 ng/ml for kortisol i fiskeplasma, for fullstendig nøyaktig
avlesing av resultat. | |
dc.description.abstract | Stress responses represent an animal’s reaction to challenging situations and can be used as a welfare indicator. Cortisol is relevant for fish welfare and is an often-measured component from the primary stress-response system. The experiments aim was to investigate if the Neogen-kit could be simplified, by replacing the extraction step with SDR from the Enzo-kit. The research question is, “Can the Neogen-kit be simplified, and at the same time keep the accurate measuring of cortisol?”. One ensured that the results from the experiment were replicable by testing the correlation between two plates. The extracted, and non-extracted showed a correlation between the plates. The next goal was to optimize the extraction method, and the extraction variant two minutes vortexing was picked as optimal. Afterward, a stress-test to test the different methods with the same sample pool of plasma before and after stress was conducted. The test implied that there was a difference in cortisol levels between before and under the stressing of the fish. The last part of the experiment tested the correlation and difference of the extracted- and non-extracted method and showed if the Neogen-procedure becomes more functional and avoids the additional charge for the Enzo-kit. It showed if it was possible to simplify the Neogen-kit and keep the original accuracy of the measuring of cortisol. The results showed a positive correlation between the methods. Because of the difference between the methods, the null hypotheses were rejected. The reasoning being that the non-extracted method had a lower cortisol concentration. Future studies should focus on finding an SDR which is more compatible with the Neogen-kit. The researchers also recommend using more samples, to get a clearer variation between the groups and avoid extreme values, which negatively affects the results. Future studies also need to find a dilution which is better suited for fish plasma cortisol, as the results need to be under 10 ng/mL to be accurate. | |
