dc.description.abstractCampylobacter jejuni og Campylobacter coli er en av de hyppigste årsakene til gastroenteritt hos pasienter med akutt diaré. Det er totalt 11 ulike Campylobacter species som er registrert som tarmpatogene, og det var derfor interessant å undersøke forekomsten av andre Campylobacter species enn Campylobacter jejuni og Campylobacter coli hos pasienter med symptomer på gastroenteritt, samt hvor stor andel av disse som er klassifisert som tarmpatogene. I dette bachelorprosjektet ble det etablert en multipleks-PCR for C. fetus og C. species for å undersøke om det å benytte primere og prober for C. fetus og C. species med målsekvensen 16S rRNA førte til deteksjon av flere C. species enn avdelingens rutineanalyse for diagnostikk av tarmpatogene agens (Seegene® AllplexTM Bacteria(I)). For å etablere multipleks-PCR for C. fetus og C. species ble analysen optimalisert, samt analysens effektivitet, spesifisitet og deteksjonsgrense bestemt. 826 pasientprøver fra pasienter med diaré ble analysert med multipleks-PCR for C. fetus og C. species, og sangersekvensering ble benyttet for å identifisere bakteriestammene som ble detektert i pasientprøvene. Resultatene viste at multipleks-PCR for C. fetus og C. species hadde god effektivitet, spesifisitet, og lav deteksjonsgrense, og kunne benyttes til påvisning av C. species i pasientprøver. Fire pasientprøver ble identifisert med sangersekvensering, hvor tre av pasientprøvene fikk identifikasjon for Campylobacter concisus, og én av pasientprøvene fikk identifikasjon for Candidatus Campylobacter infans strain. Det ble konkludert med at multipleks-PCR for C. fetus og C. species var egnet til påvisning av C. fetus og C. species i avføringsprøver fra pasienter med akutt diaré, samt at analysen detekterte flere C. species enn avdelingens rutineanalyse. Bakterieartene som ble funnet i pasientprøvene er ikke klassifisert som tarmpatogene, men funn i tidligere studier indikerer likevel at artene kan være tarmpatogene.
dc.description.abstractCampylobacter jejuni and Campylobacter coli are known to be one of the most frequent causes to gastroenteritis worldwide for patients with acute diarrhea. There are in total eleven different Campylobacter species that is known to be gastrointestinal pathogen, and therefore it is interesting to investigate the instance of other Campylobacter species than Campylobacter jejuni and Campylobacter coli in patients with symptoms of gastroenteritis, including the percentage of gastrointestinal pathogen Campylobacter species detected in patient stool samples. In this bachelor thesis, a multiplex-PCR for C. fetus and C. species was established to review if the use of primers and probes for C. fetus and C. species with the target sequence 16S rRNA, lead to detection of further C. species than the routine multiplex-PCR Allplex Bacteria (I), which is currently used for detection of gastrointestinal pathogens at the clinical department of medical microbiology at St. Olavs Hospital. To establish the multiplex-PCR for C. fetus and C. species, the analysis was optimized, additionally to the determination of the analysis`s efficiency, specificity, and detection limit. 826 stool samples from patients with diarrhea were analyzed with the multiplex-PCR for C. fetus and C. species. Sanger sequencing was practiced for the identification of bacteria detected in patient stool samples with the multiplex-PCR for C. fetus and C. species. The results affirmed that the multiplex-PCR for C. fetus and C. species had acceptable efficiency, specificity, and a low detection limit, and could be used for detection of C. species in patient samples. Four of 826 patient samples were identified with sanger sequencing, where three of the patient samples were identified as Campylobacter concisus, and one of the patient samples was identified as Candidatus Campylobacter infans strain. In conclusion, the multiplex-PCR for C. fetus and C. species was suitable for detection of C. fetus and C. species in stool samples from patients with acute diarrhea, and the analysis detected further C. species than the routine Allplex-PCR. The bacteria species that were detected in the patient samples is not classified as gastrointestinal pathogens, but newer studies indicate that they might be a cause to gastroenteritis.
