dc.description.abstractAntibiotika resistente bakterier er et stort problem i dag og kommer til å fortsette å være et stort problem i fremtiden. Det er estimert at i 2050 kommer antibiotika resistente patogener til å drepe flere folk enn kreft. Meticillin resistent Staphylococcus aureus (MRSA) er en mangfoldig resistent gram-positiv bakteriell patogen som fører til rundt 10 000 døde hvert år. MRSA sin resistans mot β-lactam antibiotika kommer fra penicillin bindende protein 2a (PBP2a). Dette enzymet, som er en del av cellevegg syntesen i bakteriene, binder ikke β-lactam antibiotika fordi det aktive setet er blokkert. Ceftaroline (1) ble funnet å bindes allosterisk til PBP2a, noe som endrer konformasjonen til enzymet, som gjør binding til det aktive setet mulig, Dette gir ceftaroline en anti-MRSA mulighet. Ceftolozane (2) binder seg ikke til det allosteriske setet og er inaktiv mot MRSA. I denne teksten skal den strukturelle grunnen for bindingen til det allosteriske setet av ceftaroline utforskes, og sammenlignet mot ceftolozane. Konklusjonen er at den svovel-bundne 1,3-tiazol ringen på C3 substituenten er den strukturelle delen som fører til at ceftaroline binder seg.
dc.description.abstractAntibiotic resistant bacteria are a big problem today and are going to continue to be a problem into the future. It is estimated that by 2050 antibiotic resistant pathogens will kill more people than cancer. Methicillin resistant Staphylococcus aureus (MRSA) is a highly resistant gram-positive bacterial pathogen that causes around 10 000 deaths annually. MRSAs resistance towards β-lactam antibiotics is caused by penicillin binding protein 2a (PBP2a). This enzyme, which is a part of cell wall synthesis in the bacteria, does not bind β-lactam antibiotics because its active site is blocked. Ceftaroline (1) was found to bind allosterically towards PBP2a, changing the conformation of the enzyme, making binding to the active site possible. This gives ceftaroline an anti-MRSA capability. Ceftolozane (2) does not bind to this allosteric seat and is inactive towards MRSA. In this thesis the structural reason for the binding to the allosteric site by ceftaroline is explored and compared to that of ceftolozane, concluding that the sulfur linked 1,3-thiazole ring substituted at C3 is the structural moiety that makes ceftaroline bind.
