dc.description.abstractI denne oppgaven utførte vi en forenklet validering av en ny metode for kvantifisering av kotinin i serum på UPLC-MS/MS. Vi undersøkte også om metoden kunne benytte en standardrekke basert på 4% bovint serumalbumin fortynnet i fosfat-buffret saltvann (BSA) i stedet for serum. Bakgrunnen for ønske om å benytte BSA var problemer med å anskaffe tilstrekkelig mengder kotininfritt serum. De ulike forsøkene ble utført ved å benytte standardløsninger og kvalitetskontroller basert på serum og BSA, samt serumprøver fra pasienter. Metoden benytter internstandardkalibrering med kotinin-D3 som internstandard og prøveopparbeidelse ble gjort av Hamilton pipetteringsrobot. Valideringen ble satt opp av ansatte ved seksjon for forskning og utvikling (FoU) ved avdeling for klinisk farmakologi. Planen omfattet undersøkelse av egnetheten for en BSA-basert standardkurve; undersøkelse av standardkurvens linearitet og måleområde, kvantifiseringsgrense, stabiliteten til retensjonstid/relativ retensjonstid, selektivitet, carry-over, presisjon, nøyaktighet, stabiliteten til forholdet mellom kvantifiseringsion og kvalifiseringsion, robusthet og IS-recovery. I tillegg ble det utført to holdbarhetsundersøkelser. Resultatene viste at alle parameterne som ble undersøkt tilfredsstilte det satte metodekravet og den BSA-baserte standardkurven hadde god riktighet. Prøvene viste seg å være holdbare fire uker på kjølerom ved 4-8 ºC og tre dager på autosampler ved 10 ºC. Det ble derfor konkludert med at metoden er god nok til å kunne benyttes til kvantifisering av kotinin i serum, og at den kan benytte PBS-baserte standarder og kvalitetskontroller.
dc.description.abstractIn this study we conducted a simplified validation of a new method for measuring serum cotinine using UPLC-MS/MS. We also examined whether this method could be validated using a calibration curve based upon standard solutions made of 4% bovine serum albumin diluted in phosphate-buffer saline (BSA) instead of using serum. The reason for using BSA was scarcity of sufficient quantities of cotinine-free serum. The laboratory experiments were performed using standard solutions and quality controls based either BSA or serum, as well as serum samples from different patients. The method applies an internal standard calibration using cotinine-D3. The sample preparation was primarily conducted using a pipetting robot called Hamilton. The validation was set up by employees at the section of Research and Development at the Department of Clinical Pharmacology. The validation included a suitability test of a standard curve based on BSA solution, an examination of the standard curve’s linearity, measuring range, a limit of quantification test, retention time / relative retention time stability, selectivity, carry-over, precision, accuracy, stability in ratio between quantification and qualification ions, robustness and a measurement of internal standard recovery. Furthermore, the validation included examination of the samples’ durability. Our results show that all the validated analytical parameters were within the requirement of this method. A BSA-based standard curve shows good accuracy when used for quantification of human serum samples. Samples could be stored at 4-8 ºC for four weeks, and at 10 ºC for three days. Based on the results of the present study we concluded that the method can be utilized for quantification of cotinine in serum, including use of BSA-based standard solutions and quality controls.
