dc.description.abstractKronisk nyresykdom (CKD) er et globalt folkehelseproblem, og utviklingen av sykdommen er assosiert med vevsinflammasjon som påvirker fibrosedannelse. Fibrose er et vanlig kjennetegn ved CKD. Sykdommen påvirker nyrestruktur og -funksjon, og vil gradvis føre til irreversible skader på organet. Dagens behandlingsalternativer verken stopper eller reverserer sykdommens utvikling. En av de største utfordringene i utviklingen av effektive behandlinger mot CKD er manglende kunnskap om de molekylære mekanismene som ligger til grunn for sykdommens utvikling. Det er stort behov for nye og bedre behandlinger for å bekjempe CKD, og utviklingen av disse avhenger av mer kunnskap om sykdommens molekylære mekanismer. For å undersøke samspillet mellom ulike molekylære mekanismer, ble det utført en undersøkelse av cytosolisk fosfolipase A2α (cPLA2α) og vitamin D reseptor (VDR) signalisering som nye terapeutiske mål for CKD. Effektene av cPLA2α-hemmeren compound 1 og den vitamin D-analoge forbindelsen calcitriol ble studert på fibroserelaterte prosesser i fibroblastcellelinjen NRK-49F. Compound 1 ble funnet å redusere levedyktigheten til TGF-β1-behandlede celler på en doseavhengig måte og redusere TGF-β1-indusert α-SMA-uttrykk. Calcitriol ble derimot ikke funnet å ha noen av disse effektene. Ved å undersøke kombinasjonen av suboptimale doser av compound 1 og calcitriol, ble kombinasjonsbehandlingen funnet å ha en bedre enn additiv effekt på TGF-β1-indusert α-SMA- og Col3a1-uttrykk. Imidlertid, ved å undersøke effekten av kombinasjonsbehandlingen på sekrert PGE2, viste det seg at den kombinerte effekten av de to forbindelsene ikke var til stede, ettersom den var lik effekten gitt av compound 1 som monoterapi. I tillegg ble den kombinerte effekten av de to forbindelsene på cellenes levedyktighet undersøkt ved en kurveforskyvningsanalyse. Resultatet fra denne analysen gav ikke et klart svar. Fokuset for denne oppgaven har vært å undersøke effekten av å kombinere cPLA2α-hemming med VDR signalisering på genuttrykk, PGE2 sekresjon og levedyktighet til NRK-49F celler. Kombinasjonen som et mulig behandlingsalternativ for CKD, kunne imidlertid verken støttes eller avvises. Derimot viste compound 1 å ha lovende effekter som monoterapi i dette CKD-modellsystemet og kan ha synergistiske effekter i kombinasjon med andre stoffer.
dc.description.abstractChronic kidney disease (CKD) is a global public health issue, and its progression is associated with tissue inflammation affecting the process of fibrogenesis. Fibrosis is a common characteristic of CKD. The disease affects kidney structure and function, progressively leading to irreversible damage to the organ. Today’s treatment options do neither stop nor reverse the disease progression. One of the main obstacles to develop effective treatments against CKD lay in the inadequate knowledge of the molecular mechanisms underlying the progression of the disease. To combat CKD there is a large need for new and better treatments and the development of these depend on the unravelling of the molecular mechanisms. To investigate the interplay between different molecular mechanisms, an investigation into cytosolic phospholipase A2α (cPLA2α) and vitamin D receptor (VDR) signaling as novel therapeutic targets in CKD, was performed. The effects of the cPLA2α inhibitor compound 1 and the vitamin D analogue calcitriol were studied on fibrosis-related processes in the fibroblast cell line NRK-49F. Compound 1 was found to reduce the viability of TGF-β1-treated cells in a dose-dependent manner and reduce the TGF-β1-induced α-SMA expression. Calcitriol, on the other hand, was not found to exert any of these effects. When investigating the combination of suboptimal doses of compound 1 and calcitriol, the combinational treatment was found to have a greater than additive effect when looking at TGF-β1-induced α-SMA and Col3a1 expression. However, when investigating the effect of the combinational treatment on secreted PGE2, the combined effect of the two compounds was shown to not be present since it matched the effect exerted by compound 1 as a monotherapy. In addition, the combination of the compounds was investigated for a synergistic effect on viability through a curve shift analysis. The result of this analysis was inconclusive. The focus of this thesis has been to investigate the effect of combining cPLA2α-inhibition with VDR signaling on gene expression, PGE2 secretion and cell viability in the NRK-49F cell line. However, the combination as a potential treatment option for CKD could neither be supported nor rejected. Compound 1, on the other hand, showed promising effects as a monotherapy in this CKD model system and may have synergistic effects with other agents.en
