dc.description.abstractFor å undersøke effekten av langvarig stress gjennom analyse av cellesyklus, ble CHSE-214 celler dyrket in vitro og stresset med bruk av varme. Optimal vekst til CHSE-214 ligger på 20 grader celcius, i stressanalysen ble brett med seks brønner plassert til varmestress på 26 grader celcius. Forsøket ble gjentatt to ganger, der fem tidspunkter ble analysert i hvert forsøk; 24-, 48-, 72-, 96- og 120 timer. Grunnet SARS-CoV-2, ble det ikke tid til å analysere 120 timer stress i forsøk 2. Etter stressinitiering ble prøvene samlet, fiksert med etanol og farget med propidium iodide (PI), før analyse av cellesyklus på flowcytometer. Varmestress på 26 grader celcius ga en endring i cellesyklus på åtte av de femten undersøkte punktene av statistisk signifikans. Endringen ble undersøkt i de tre fasene G1/G0-, S-, og G2/M på alle de fem prøvetidspunktene. Resultatene kan være påvirket av forhastet stressinitiering i forsøk 2 som har resultert i store variasjoner mellom parallellene. De stressede cellene viste en nedgang i G1/G0-fasen mot kontrollceller dyrket på 20 grader celsius, der nedgangen økte med lengden påført stress. Det ble også observert en nedgang i G2/M-fasen mot kontroll. Derimot viste de stressede cellene en økning i andelen celler i S-fasen mot kontroll som kan indikere en S-fase cellesyklusarrest.
dc.description.abstractTo investigate the effect of long-term stress through cell cycle analysis, CHSE-214 cells were cultured in vitro and stressed using heat. The optimum growth of CHSE-214 is at 20 degrees Celsius. Trays with six wells were placed at a temperature of 26 degrees Celsius in the stress analysis. The experiment was repeated twice, with five different time intervals analysed in each experiment; 24-, 48-, 72-, 96-, and 120 hours. As a result of SARS-CoV-2, there was no time to analyse 120 hours of stress in experiment 2. Following the initiation of stress, the samples were collected, ethanol-fixed and propidium iodide (PI) stained, before the cell cycle on the flow cytometer was analysed. Heat stress of 26 degrees Celsius caused a change in the cell cycle in eight out of the fifteen statistically significant points examined. The change was examined in the three phases G1/G0, S, and G2/M, in all five time intervals. The results may have been influenced by the rushed stress initiation in Experiment 2, which resulted in large variations between the parallels. Stress cells showed a decrease in the G1/G0 phase towards control cells grown at 20 degrees Celsius, where the decrease increased with the duration of stress applied. A decrease in the G2/M phase towards control has also been observed. The stress cells, on the other hand, showed an increase in the proportion of cells in the S-phase to control, which may indicate an S-phase cell cycle arrest.
