dc.description.abstractMålet for denne masteroppgaven er å utvikle en analyseprotokoll for statistisk analyse av en biomarkør som en funksjon av kliniske og medisinske data for pasienter med de autoimmune sykdommene systemic lupus erythematosus, rheumatoid arthritis og seronegative rheumatoid arthritis før, under og etter svangerskapet. I denne oppgaven vil vi fokusere på biomarkøren Lipocalin2. Dataene inneholder repeterte målinger for 39 svangerskap som har målinger på opptil syv ulike tidspunkt. Sammenhengen mellom Lipocalin2 og de kliniske og medisinske dataene blir modellert ved bruk av lineære miksede modeller. Gjennom oppgaven fokuserer vi på valg av en god regresjonsmodell ved bruk av såkalt opp-ned modell-seleksjonstrategi med AIC (referert til som asymptotisk AIC) og bootstrap AIC som bestemmelseskriterium. Videre ser vi på bruk av bootstrapping til å lage konfidensintervaller for parameterne i modellen og sammenligner disse med asymptotiske konfidensintervaller. Vi bruker fire ulike bootstrapmetoder, en ikke-parametrisk, to parametriske og en semi-parametrisk. Alle disse med unntak av den ikke-parametriske metoden, betegnet paret bootstrap, utfører versjoner av residual bootstrapping. Bootstraputvalget trekkes enten fra den empiriske fordelingen eller fra normalfordelingen. Den best tilpassede lineære miksede modellen for Lipocalin2 inkluderer sykdom, BMI og et tredjegrads polynom i tid som faste effekter. De tilfeldige effektene består av skjæringspunkt for svangerskap og stigningstall i tid. Modellen er valgt basert på asymptotisk AIC. Videre ser vi at asymptotisk AIC og bootstrap AIC velger ulike beste regresjonsmodeller. Vi observerer også at de asymptotiske konfidensintervallene stemmer godt overens med konfidensintervallene som lages ved hjelp av bootstrap metoder.
dc.description.abstractThe aim of this thesis is to set up a pipeline for a statistical analysis of one biomarker as a function of clinical and medical data for patients with the autoimmune diseases systemic lupus erythematosus, rheumatoid arthritis and seronegative rheumatoid arthritis before, during and after pregnancy. The biomarker of interest in this thesis is the protein Lipocalin2. We work with a dataset which consists of repeated measurements for 39 different pregnancies with measurements at up to seven different timepoints. The relationship between Lipocalin2 and the clinical and medical data is modeled using linear mixed models. Throughout the thesis we focus on the selection of an appropriate regression model using the top-down model selection strategy with AIC (referred to as asymptotic AIC) and bootstrap AIC as selection criterion. Furthermore, we look at the use of bootstrapping to create confidence intervals for the parameters and compare these to asymptotic confidence intervals. We use four different bootstrap methods, one nonparametric, two parametric and one semi-parametric. All the methods except the nonparametric, denoted paired bootstrap, perform variants of residual bootstrapping. The bootstrap samples are either drawn from the empirical distribution or the normal distribution. The selected best linear mixed model for Lipocalin2 includes disease, BMI and a third degree polynomial in time as fixed effects. The random part consists of a random intercept for pregnancy and a random slope in time. This is the model which is selected by the asymptotic AIC. Furthermore, we see that the asymptotic AIC and the bootstrap AIC select different models. We also observe that the asymptotic confidence intervals agrees well with the ones created using bootstrap methods.
