dc.description.abstractIntroduksjon: Fysisk inaktivitet er selvstendig forbundet med flere kroniske sykdommer og redusert levetid. For å begrense disse negative effektene, har fysisk aktivitetsanbefalinger blitt fremmet for å øke fysisk aktivitet og redusere fysisk inaktivitet. På tross av dette er forekomsten av fysisk inaktivitet uendret på verdensbasis. Arv har vist seg å påvirke fysiske aktivitetsnivåer, og genetisk variasjon foreslås som en forklaring på hvorfor noen velger å være fysisk aktive, og andre fysisk inaktiv. Formål: Å identifisere genetiske variasjon assosiert med forskjellige fenotyper av fysisk inaktivitet ved å bruke genomvide assosiasjonsanalyser. I tillegg skal vi undersøke hvilke gener som mulig endres med denne variasjonen, hvor de uttrykkes og hvilke biologiske funksjoner som kan forandres. Metode: Fire ulike fenotyper av fysisk inaktivitet ble definert ved hjelp av spørreskjemaer fra HUNT-undersøkelsen: 1) Fysisk aktivitetsanbefalinger (FAA) sammenlignet personer som gjorde mindre enn 30 minutter fysisk aktivitet hver dag (11,040 tilfeller) med de som gjorde 30 minutter eller mer (36,420 kontroller). 2) Fysisk inaktivitet på fritiden (FIF) sammenlignet 13,550 fysisk inaktive tilfeller med 24,149 svært fysisk aktive kontroller. 3) Sitte tid (SIT) var basert på gjennomsnittlig tid sittende hver dag (n=57,901). 4) Fysisk aktivitetsscore (FAS) basert på fysisk aktivitetsfrekvens, varighet og intensitet (n=47,470). Fenotypene ble analysert med logistiske og lineære miksede modeller, undersøkt for linket nedarving (LN), nærliggende kandidatgener ble kartlagt, og hvilke vev genene uttrykkes i ble analysert. Kovariater ble justert for trinnvis i tre modeller. Et søk på vanlig variasjon med mindre allel frekvens (MAF) på >0.01 og ett søk etter sjeldne genetiske variasjon med MAF mellom 0.01 og 0.001 ble utført. Resultater: For vanlig genetisk variasjon fant vi et signifikant lokus på kromosom 8 for fenotypen FAA. Her var rs73183891 den mest signifikante genetiske markøren (p=3,5x10^-8) over signifikansgrensen på 5x10^-8, og hadde 32 andre genetiske markører med LN. Det nærmeste kartlagte genet var CSMD1, som uttrykkes i sentralnervesystemet og primært pannelappen. På jakt etter sjeldne genetisk variasjon var to nye HUNT-spesifikke genetiske markører lokalisert på kromosom 4 når vi undersøkte FIF fenotypen. Konklusjon: Vi fant vanlig genetisk variasjon på kromosom 8 med CSMD1 som nærmeste gen assosiert med å gjøre mindre enn 30 minutter fysisk aktivitet daglig. Gen som transkriberes i pannelappen har tidligere vært forbundet med fenotyper av fysisk aktivitet, og nå også fysisk inaktivitet. Vi fant også sjeldne HUNT-spesifikke genetiske variasjon som er assosiert med fysisk inaktivitet på fritiden.
dc.description.abstractIntroduction: Physical inactivity has independently been associated with increased risk of several chronic diseases and reduced longevity. To constrain this adverse effect, physical health recommendations have been advocated to increase physical activity and reduce physical inactivity. Albeit this effort, the worldwide prevalence of physical inactivity is not reduced. Increased knowledge of factors associated with physical activity habits is therefore essential. Heritage have been shown to affect physical activity levels, and genetic variation are suggested to explain why some are active, and others not. Aim: To identify genetic variants associated with different physical inactivity phenotypes using genome-wide association analyses. We will also explore which genes may be affected by the genetic variation, where these genes are expressed, and which biologic functions might be affected. Method: Four different phenotypes of physical inactivity was defined using questionnaires from the HUNT survey: 1) Physical activity recommendations (PAR) compared people who did less than 30 minutes physical activity each day (11,040 cases) with those who did 30 minutes or more (36,420 controls). 2) Leisure time physical inactivity (LPI) compared 13,550 physical inactive cases with 24,149 highly physical active controls. 3) Sitting time (SIT) based on average time sitting each day (n=57,901). 4) Physical activity score (PAS) based on physical activity frequency, duration and intensity (n=47,470). The phenotypes were analysed with logistic and linear mixed models, checked for linkage disequilibrium (LD) structures, nearby candidate genes were mapped, and tissue-specific expressions was analysed. Covariates was adjusted for stepwise in three models. One search for common genetic variation with minor allele frequency (MAF) of >0.01 and one search for rare genetic variation with MAF between 0.01 and 0.001 was performed. Results: For common genetic variation, one significant locus on chromosome 8 was located when investigating PAR. In this locus, rs73183891 was the lead short nucleotide polymorphism (SNP) with a p-value of 3.5x10^-8 which is below the significance threshold of 5x10^-8, and had 32 other SNPs in LD. The closest mapped gene was CSMD1, which is expressed in the central nervous system, primarily the frontal cortex. In search of rare genetic variation two novel HUNT specific SNPs was located on chromosome 4 when investigating the LPI phenotype. Conclusion: We found common genetic variation with CSMD1 as the closest gene on chromosome 8 associated with doing less than 30 minutes of physical activity each day in a Norwegian population. Genes transcribed in the frontal cortex have previously been associated with phenotypes of physical activity, and now also physical inactivity. Rare HUNT specific genetic variation was discovered and associated with leisure time physical inactivity.
