dc.description.abstractMålet med denne masteroppgaven var å undersøke effektene av de ulike reagensene brukt i syntesen av enantiomert rene forløpere, som et bidrag til økt miljøvennlighet i totalsyntesene av (S)-atenolol og (S)-metoprolol. Syntesen av forløperen 4-(3-klor-2-hydroksypropoksy)bensenacetamid (1b) begynte med å reagere utgangsstoff 2-(4-hydroksyfenyl)acetamid (1a) med natrium hydroksid og epiklorhydrin. Dette ga 1b, 4-(2-oksiran-2-ylmetoksy)bensenacetamid (1c), samt dimeren 2,2’-(((2-hydroksypropan-1,3-diyl)bis(oksy))bis(4,1-fenylen)diacetamid (1d). I denne masteroppgaven ble effekten av ulike ekvivalenter natriumhydroksid og epiklorhydrin i tillegg til ulike reaksjonstider studert. Undersøkelsene antydet at dimer 1d er et termodynamisk favorisert produkt som kan unngås ved reduserte reaksjonstider. Undersøkelsene indikerte videre at katalytiske ekvivalenter av natriumhydroksid reduserer dannelsen av dimer 1d. Full omsetning av utgangsstoffet 1a var mulig ved bruk av katalytiske mengder natriumhydroksid, noe som antyder at reaksjonen er spesifikk-base katalysert. Undersøkelsene viste også at økte ekvivalenter epiklorhydrin økte reaksjonshastigheten, noe som indikerer at reaksjonen følger standard SN2-reaksjonskinetikk. Til slutt viste studiene at epoksid 1c er termodynamisk favorisert ved basiske betingelser, og at ved katalytiske ekvivalenter natriumhydroksid vil kloralkohol 1b favoriseres over epoksid 1c. Syntesen av ren kloralkohol ble gjennomført ved syrekatalysert ringåpning av epoksid 1c. Både litiumklorid sammen med eddiksyre og saltsyre ble forsøkt. Undersøkelser viste at å øke ekvivalentene av eddiksyre over fem ekvivalenter ikke økte reaksjonshastigheten ytterligere. Å bruke to ekvivalenter litiumklorid sammenlignet med én, økte reaksjonshastigheten noe som indikerer at reaksjonen følger standard SN2-reaksjonskinteikk. Ved å bruke tre ekvivalenter litiumklorid ble en redusert reaksjonshastighet observert. Samme reaksjon gjennomført med saltsyre i stedet for litiumklorid og eddiksyre viste ingen forandringer i omsetningshastighet. Kinetisk oppløsning av kloralkohol 1b ble gjennomført med to ulike preparater av Candida antarctica Lipase B (CALB), ett fra SyncoZymes og ett fra Sigma Aldrich, for å undersøke preparatenes forskjeller i aktivitet og enantioselektivitet. CALB-preparatet fra SyncoZymes viste omtrent dobbelt så høy aktivitet for dannelsen av (R)-1b og ester (S)-1-(4-(2-amino-2-oksoetyl)fenoksy)-3-klorpropan-2-yl butyrat ((S)-1e), sammenlignet med preparatet fra Sigma Aldrich. Den kinetiske oppløsningen av kloralkohol 1b med CALB fra SyncoZymes viste ufullstendig omsetning av (S)-1b etter 24 timer, med e.e. = 86%. Da ingen av de kinetiske oppløsningene hadde gått til fullstendig omsetning etter 24 timer, var det ikke mulig å definere forskjellene i enantioselektivitet. Reaksjonen mellom 4-(2-metoksi)fenol (2a) og epiklorhydrin under basiske betingelser gav 1-klor-3-(4-(2-metoksyetyl)fenoksy)propan-2-ol (2b) og 2-((4-(2-metoksyetyl)fenoksy)metyl)oksiran (2c), samt urenhetene A, B og D. I denne masteroppgaven ble effektene av ulike ekvivalenter natriumhydroksid og epiklorhydrin, samt effekten av temperatur og reaksjonstider undersøkt. Undersøkelsene viste at reaksjonen gikk til fullstendig omsetning av 1a ved katalytiske ekvivalenter natriumhydroksid, noe som indikerte at reaksjonen var spesifikk-base katalysert. Ved å bruke katalytiske mengder natriumhydroksid og et overskudd av epiklorhydrin var det mulig å unngå dannelsen av urenhetene. Videre viste undersøkelsene at epoksid 1c var det favoriserte produktet, med mindre reaksjonene ble gjennomført ved 80°C noe som favoriserte kloralkohol 2b. Å gjennomføre reaksjonene ved høyere temperatur førte også til høyrere dannelse av urenhetene, noe som indikerte at urenhetene var kinetisk foretrukket. For å danne ren kloralkohol 2b ble råproduktet fra reaksjonen reagert med litiumklorid og eddiksyre, i en syrekatalysert ringåpning av epoksid 2c. Kinetisk oppløsning av klorlakohol 2b ble gjennomført på samme måte som for kloralkohol 1b, med de samme preparatene av CALB. De kinetiske oppløsningene viste at CALB hadde høyere aktivitet i kinetisk oppløsning av 2b sammenlignet med 1b. CALB-preparatet fra SyncoZymes hadde viste omtrent dobbelt så høy aktivitet som CALB-preparatet fra Sigma Aldrich, og dannet (R)-2b i 100% e.e. etter 1 time reaksjon med en høyere enantioselektivitet sammenlignet med CALB-preparatet fra Sigma Aldrich.
dc.description.abstractThe aim of this thesis was to gain a better understanding regarding the impacts of reagents used in the synthesis of precursors for the total synthesis of (S)-atenolol and (S)-metoprolol as a contribute to making their synthesis more environmentally friendly. The synthesis of (S)-atenolol precursors begun with deprotonation of 2-(4-hydroxyphenyl)acetamide (1a) by sodium hydroxide followed by a nucleophilic attack on epichlorohydrin in a SN2-manner forming 4-(3-chloro-2-hydroxypropoxy)benzeneacetamide (1b) and 4-(2-oxiran-2-ylmethoxy)benzeneacetamide (1c) with 2,2’-(((2-hydroxypropane-1,3-diyl)bis(oxy))bis(4,1-phenylene)diacetamide (1d) as biproduct. By variating equivalents of both sodium hydroxide and epichlorohydrin as well as reaction times studies showed that the formation of dimer 1d might be a thermodynamically favored product, thus avoided by reducing the reaction time. Formation of 1d could be minimized by decreasing the equivalents of sodium hydroxide used in the reaction. Studies also showed that the reaction proceeded to full conversion of starting material 1a using 0.3 equivalents of sodium hydroxide which indicates that the reaction was a matter of specific-base catalysis. Increased equivalents of epichlorohydrin increased the reaction rate, indicating that the reaction follows a standard SN2-reaction mechanism. Investigations also suggested that epoxide 1c is the thermodynamically favored product, while chloroalcohol 1b is the kinetically favored one under basic conditions. Thus, by decreasing the equivalents of sodium hydroxide, the equilibria between epoxide 1c and chloroalcohol 1b would shift to where 1b was the major product of the reaction. The chloroalcohol 1b were used in the enantiodifferentiating reactions later in this thesis and were obtained pure when a mixture of 1b and epoxide 1c were reacted with either lithium chloride and acetic acid or hydrochloric acid. By changing the equivalents of lithium chloride and acetic acid, studies reviled that exceeding 5 equivalents of acetic acid did not increase the reaction rate. Further, studies showed that using 2 equivalents of lithium chloride increased the reaction rate compared to using 1 equivalent. Using 3 equivalents of lithium chloride decreased the reaction rate. Using hydrochloric acid did not change conversion rates of epoxide 1c into chloroalcohol 1b compared to using lithium chloride and acetic acid. The enantiodifferentiating reactions were performed as a kinetic resolution of chloroalcohol 1b forming (S)-1-(4-(2-amino-2-oxoethyl)phenoxy)-3-chloropropan-2-yl butyrate ((S)-1e).Kinetic resolution of chloroalcohol 1b were performed with two different preparations of Candida antarctica Lipase B (CALB), one from SyncoZymes and one from Sigma Aldrich in order to compare their activities and enantioselectivities. Studies showed that the activity of the CALB preparation from SyncoZymes was about twice as high as the CALB preparations from Sigma Aldrich. Neither reactions proceeded to full conversion of (S)-1b within 24 hours. Because of the differences in conversion reached within the 24 hours, it was not possible to state which preparation that showed the highest enantioselectivity. As substrate for kinetic resolution of (S)-metoprolol precursor 1-chloro-3-(4-(2-methoxyethyl)phenoxy)propan-2-ol (2b) were synthesized when starting material 4-(2-methoxyethyl)phenol (1a) performed a nucleophilic attack on epichlorohydrin under basic conditions. This gave both 2b and 2-((4-(2-methoxyethyl)phenoxy)methyl)oxirane (2c) along with impurities A, B and D. The impact of equivalents of sodium hydroxide as well as temperatures and reaction time were investigated. Studies performed on this reaction showed that it was possible to achieve full conversion of starting material 2a by using catalytical amounts of sodium hydroxide, thus indicating that this reaction is specific-base catalyzed. Studies reviled that using catalytic equivalents of base and excessive equivalents of epichlorohydrin avoided formation of impurities. Under basic conditions the formation of epoxide 2c seemed to be favored over chloroalcohol 2b, but when increasing the temperature to 80°C the favored product was chloroalcohol 2b. To synthesize pure chloroalcohol 2b, crude products obtained from the reaction given in Scheme 0.4 were reacted with lithium chloride and acetic acid. The studies performed showed that the impurity present also react with lithium chloride and acetic acid. Purification prior to this reaction is therefore crucial. Using 1 equivalent of lithium chloride and 5 equivalents of acetic acid was enough for this reaction to proceed to full conversion of epoxide 2c if no impurities were present. Kinetic resolution of chloroalcohol 2b was performed with the same CALB preparations as used for the kinetic resolution of 1b. The activities of the enzymes for the conversion of chloroalcohol 2b into enantiomerically enriched (R)-2b and ester (S)-2e were higher for the metoprolol precursors than for atenolol. The CALB preparation from SyncoZymes showed full conversion of (S)-2b after 1 hour of reaction to (R)-2b in 100% e.e., see, with a higher enantioselectivity towards (S)-2b than CALB from Sigma Aldrich.
