dc.description.abstractOppgaven tar for seg optimalisering av en metode for å påvise fagocytose i leukocytter ved hjelp av flowcytometri. Fagocytose er en prosess der en celle omslutter og tar opp større partikler for å deretter bryte dem ned. Dette er en viktig del av den immunologiske funksjonen til flere typer leukocytter. For å påvise fagocytose ble pHrodo Green E. coli Bioparticles Conjugate fra Thermofisher brukt. Konjugatet skal emittere sterkere fluorescens ved lav pH, som er optimalt ved påvisning av fagocytose. Anti-CD45 ble brukt til å få over oversikt over de ulike leukocyttpopulasjonene. Anti-CD16 ble brukt for å nærmere studere de profesjonelle fagocyttene. Tidligere forskning og vitenskapelige artikler om påvisning av fagocytose og overflateantigen ble brukt for å samle inn informasjon. Basert på anbefalinger fra produsent og tidligere forsøk ble reagenser og utstyr valgt ut. Enkelte justeringer ble gjort på grunnlag av hva som var tilgjengelig. En midlertidig prosedyre ble utarbeidet og tilpasset etter hvert som ulike punkter i prosedyren ble utprøvd. Det ble observert resultater som stemte overens med at konjugatet hadde blitt fagocyttert. Disse resultatene ble forbedret ved å justere de ulike parameterene. Resultatene ble imidlertid påvirket av at fritt konjugat emitterte betydelig høyere fluorescens enn forventet. Det var bare mulig å skille fritt konjugat fra antatt fagocyttert konjugat ved bruk av anti-CD45. Det ble forsøkt å benytte Trypan Blue som quencher, men dette var ikke vellykket. Etter utprøving og tilpasning av prosedyren ble en endelig prosedyre utarbeidet. Prosedyren kan brukes for å påvise fagocytose i leukocytter ved hjelp av flowcytometri, men metoden bør kontrolleres for å bekrefte at konjugatet har blitt fagocyttert.
dc.description.abstractThis thesis covers optimization of a method for detecting phagocytosis in leukocytes by utilizing flow cytometry. Phagocytosis is a process where a cell envelope and absorbs bigger particles in order to break them down. This is an important part of the immunological function of several types of leukocytes. pHrodo Green E. coli Bioparticles Conjugate from Thermofisher was utilized in order to detect phagocytosis. The conjugate is supposed to emit stronger fluorescence when exposed to low pH, which is ideal for detecting phagocytosis. Anti-CD45 was used in order to obtain an overview of the different leukocyte-populations. Anti-CD16 was used in order to further study the professional phagocytes. Earlier studies and scientific articles about the detection and measurement of phagocytosis were used to collect information about the topic. Reagents and equipment were chosen based on recommendations from the manufacturer. Certain changes were made based on availability and accessibility. A temporary procedure was created and adjusted as different parts of the method were tested. After the first run, results suggesting that the conjugate had been phagocytosed were observed. These results were improved by adjustments made to the different parameters of the procedure. However, the results were also affected by the fact that free conjugate emitted a higher fluorescens than expected. The only way to separate the free conjugate from the conjugate assumed to have been phagocytosed was through the use of anti-CD45. There were attempts at using Trypan Blue as a quenching buffer, but this was not successful. The trials and adjustments of the assay has resulted in a finished procedure. This procedure can be utilized for the detection of phagocytosis in leukocytes by using flow cytometry, but the method should be validated in order to confirm that the conjugate has been phagocytosed.
