Effect of natural astaxanthin sources on pigmentation and related biochemical mechanisms in Salmo salar L.

Abstract
Atlantisk laks (Salmo salar L.) har en fremtredende rødfarge som er et resultat av avsetning av karotenoider i muskelvev, derav spesielt astaxanthin. Tilsetning av pigment i fôret til oppdrettslaks er viktig ettersom fargen på kjøttet i salmonider er en viktig kvalitetsparameter og lipid perokisdasjon påvirker den helhetlige kvaliteten. I løpet av de siste årene har det også blitt mer populært å se etter grønnere og mer bærekraftige løsninger for erstatte bruken av syntetisk astaxanthin, hovedsaklig brukt i dag. Dermed er det av høy interesse å studere effekten av astaxanthin fra naturlige kilder på de nevnte kvalitetsparametrene. Målet for denne avhandlingen var derfor å undersøke effektiviteten av astaxanthin i atlantisk laks fôret med astaxanthin fra de ulike naturlige kildene algen Haematococcus pluvialis, gjæren Pfaffia rhodozyma, og bakterien Paracoccus carotinifaciens i form av farge, lipidprofil, oksidasjonsstatus, og genuttrykk sammenlignet med laks fôret syntetisk astaxanthin.
Denne avhandlingen var en del av et større forskningsprosjekt i samarbeid med BioMar AS. Fôringseksperimentet ble utført i Hirtshals, Danmark og startet 02.11.22 og endte 31.01.23. I denne avhandlingen ble fire ulike dietter med fire ulike astaxanthin kilder vurdert, H. pluvialis, P. rhodozyma, P. carotinifaciens, og syntetisk astaxanthin, hvor fiskene av hver diett (n = 93 per fôr) ble fordelt på tre tanker (n = 31 per tank). De kolorimetriske parametrene og karatenoidprofilen ble målt i muskelregionen Norsk kvalitetskutt (NQC) ved bruk av henholdsvis DigiEye og HPLC. Oksidasjonsstatus ble også undersøkt ved å analysere lipidprofilen i NQC regionen samt ratioen av glutation (GSH)/glutationdisulfid (GSSG) i både NQC og lever. Analyse av genene nrf2.1, keap1b1, hsp70, og H2Q8 ble utført i muskelvev og pyloric caeca (PC) for å undersøke sammenhengen mellom genuttrykk og oksidasjonsstatus. Signifikante forskjeller mellom fisk fôret ulike astaxanthinkilder ble anlaysert ved bruk av enveis variansanalyse (ANOVA), Tukeys ærlig signifikant forskjell (HSD), uavhengig T-test, og den ikke-parametriske testen Kruskal-Wallis.
Resultatene viste at astaxanthin fra de naturlige kildene H. pluvialis, P. rhodozyma, og P. carotinifaciens var like effektiv som syntetisk astaxantin i form av visuell oppfatning, oksidasjonsstatus, og lipidperoksidasjon ettersom ingen signfikante forskjeller (p > 0.05) i a* verdi, GSH/GSSG ratio, eller lipidprofil, ble funnet. Videre var det ingen signifikante forskjeller i utrykk av genene nrf2.1, keap1b1, og hsp70, hverken i muskelvev eller i PC. Laks fôret syntetisk astaxanthin viste derimot høyere konsentrasjon av total astaxanthin i muskelvev sammenlignet med laks fôret H. pluvialis og P. carotinifaciens. Fisk fôret med astaxanthin fra P. rhodozyma viste de mest fordelaktige resultatene da denne gruppen hadde numerisk høyere a*-, b*-, og C* verdier, samt lavest L* verdi. Det var heller ingen signifikante forskjeller i konsentrasjon av total astaxanthin i muskelvev mellom gruppene fôret P. rhodozyma og syntetisk astaxanthin. I henhold til pigmentering og visuell oppfatning kan dermed P. rhodozyma bli ansett som en potent konkurrent for syntetisk astaxanthin. Avslutningsvis kan det konkluderes med at astaxanthin fra de ulike naturlige kildene H. pluvialis, P. rhodozyma, og P. carotinifaciens viste seg å være like effektiv som syntetisk astaxanthin i henhold til farge, lipidprofil, oksidasjonsstatus og genuttrykk. Atlantic salmon (Salmo salar L.) has a prominent red flesh color, caused by the deposition of carotenoids, especially astaxanthin. Considering that the color of the salmonid flesh is an important quality parameter and that lipid peroxidation influence the overall quality, the addition of pigments to the feed of farmed salmon is of high importance. In recent years it has also become increasingly more popular to look for greener and more sustainable solutions to replace the use of synthetic astaxanthin, predominantly used today. Considering this, it is of high interest to study the effect of astaxanthin from natural sources on the aforementioned quality parameters. Therefore, the aim of the present thesis was to investigate the efficiency of astaxanthin in Atlantic salmon fed astaxanthin from different natural sources, namely the algae Haematococcus pluvialis, the yeast Pfaffia rhodozyma, and the bacteria Paracoccus carotinifaciens in terms of coloration, astaxanthin deposition, lipid profile, oxidation status, and genomic expression compared to salmon fed synthetic astaxanthin.
The present thesis was conducted in collaboration with BioMar AS and was a part of a larger research project. The feeding experiment was conducted in Hirtshals, Denmark and started 02.11.22 and ended 31.01.23. In the present thesis, four diets with four different astaxanthin sources, H. pluvialis, P. rhodozyma, P. carotinifaciens, and synthetic astaxanthin, were considered, where the fish fed each diet (n = 93 per feed) were distributed over three tanks (n = 31 per tank). Colorimetric parameters and the carotenoid profile were measured in the Norwegian Quality Cut (NQC) region of the muscle tissue using DigiEye and HPLC, respectively. Additionally, the oxidation status was investigated by analysing the lipid profile in the NQC and the ratio of glutathione (GSH)/glutathione disulfide (GSSG) in both the NQC and liver. Genomic analyses of the genes nrf2.1, keap1b1, hsp70, and H2Q8 were performed in the muscle tissue and pyloric caeca (PC) to investigate the connection between gene expression and oxidation status. The significant differences between the fish fed feed supplemented with astaxanthin from the different sources were analyzed using one-way analysis of variance (ANOVA), Tukey's honest significant difference (HSD), the independent T-test and the non-parametric test Kruskal-Wallis.
The results showed that astaxanthin from the natural sources H. pluvialis, P. rhodozyma, and P. carotinifaciens were as efficient as synthetic astaxanthin in terms of visual perception, oxidation status, and lipid peroxidation as there were no significant differences (p > 0.05) in a* value, GSH/GSSG ratio, or lipid profile. Additionally, there were no significant differences in genomic expression of nrf2.1, keap1b1, or hsp70, in neither the muscle tissue nor the PC. However, the fish fed synthetic astaxanthin had a significantly higher concentration of total astaxanthin in the flesh compared to fish fed H. pluvialis and P. carotinifaciens. The fish fed P. rhodozyma showed the most favorable results as it had the numerically highest a*-, b*-, and C* values, and the lowest L* value. Moreover, no significant difference in total muscle astaxanthin concentration was observed between the groups fed P. rhodozyma and synthetic astaxanthin. Therefore, P. rhodozyma can be considered as a potent competitor to synthetic astaxanthin in terms of flesh pigmentation. In conclusion, astaxanthin from the natural sources H. pluvialis, P. rhodozyma, and P. carotinifaciens in regards to coloration, lipid profile, oxidation status, and genomic expression has been shown to be as efficient as synthetic astaxanthin.
