Abstract
Akvakulturindustrien vokser, og behovet for oppdrettsfisk er anslått å ha en årligøkning på 5.6%. Fordi innholdet av marine ingredienser i fôret has sunket betraktelig siden nittitallet, på grunn av naturlige begrensninger i fangsten av villfisk, erdet et behov for bærekraftige alternativer. Thraustochytrider er heterotrofe, obligat marine protister som akkumulerer store mengder ω-3 flerumettede fettsyrerved bruk av et polyketidsyntase-lignende system.Mye forskning har blitt gjort på mediebetingelser, men kunnskapen om produksjonen av lipider er begrenset. Ved å undersøke metabolomet og lipidomet tilthraustochytrider kan nye muligheter for optimalisering av lipidproduksjonen åpneseg.Tre bioreaktoreksperimenter ble gjort, der alle hadde kontinuerlig måling av pHog CO2 i utslippsgassen. Det første eksperimentet ble designet som en referanse,og de to andre eksperimentene ble designet for å undersøke hvordan mangelen påtilgjengelig O2 påvirket lipidproduksjonen. Referanseeksperimentet resulterte i enproduksjon av 15.59 ± 0.78% g/L tørrvekt, med et lipidinnhold på 61.78 ± 2.85%.Triacylglycerol var den største lipidklassen (>90%), etterfulgt av fosfatidylglycerolog lysofosfatidylcholine. Innholdet av DHA var på det meste 27.1% av de totalefettsyrene, og sank til 18.6% ved slutten av eksperimentet. En økning i konsentrasjonen av et utvalg metabolitter ble observert under lipidakkumuleringsfasen.NADPH var den eneste reduksjonsagenten som økte i konsentrasjon over tid, noesom indikerte at NADPH ikke var en begrensende faktor for lipidproduksjon. T66 og kan være medvirkende til å optimalisere organismens lipidproduksjon. The aquaculture industry is blooming, and the demand for farmed fish will havean annual growth of 5.6%. As the content of marine ingredients in the feed hasdecreased significantly, due to the natural limitations in the supply of wild caughtfish, sustainable alternatives are needed. Thraustochytrids are heterotrophic, obligate marine protists that accumulate high quantities of ω-3 polyunsaturated fattyacids (PUFAs) using a polyketide synthase-like pathway.A lot of research has been done on media requirements, but the knowledgeregarding the production of PUFAs is limited. Investigating the metabolome andlipidome of thraustochytrids could open up new possibilities for optimization ofPUFA production in thraustochytrids.Three batch bioreactor experiments were performed, the first experiment wasdesigned as a reference, and the two other experiments were designed to investigate how decreasing the availability of O2 would affect the lipid production. Thereference experiment resulted in a production of 15.59 ± 0.78% g/L cell dry weight(CDW), with a lipid content of 61.78 ± 2.85%. Triacylglycerol was the most abundant lipid class (>90%), followed by phosphatidylglycerol and lysophosphatidylcholine. The content of DHA was 27.1% of total fatty acids (TFA) at the most,and decreased to 18.6% of TFA at the end of the experiment. An increase in theconcentration of certain metabolites was found to occur in the lipid accumulationphase. It was found that reducing the availability of O2 hours after nitrogen limitation found place resulted in a stable culture, whose production of CDW andlipids did not deviate much from the references.These results contribute to the understanding of the metabolism of Aurantiochytrium sp. T66 and may contribute to optimizing the organism as a productionorganism.
