dc.description.abstract | Plantecellers cellevegg fungerer som strukturell støtte og beskytter cellene mot miljøstress.
Celleveggens integritet (CWI) opprettholdes av en dedikert mekanisme for celleveggintegritetsvedlikehold (CWIM) og bedre forståelse av denne prosessen kan bidra til forbedring av
avlinger til mat og bioenergi produksjon. Her ble 30 T-DNA insersjonslinjer, for 24
kandidatgener muligens involvert i CWI basert på tidligere transkriptomikkdata, genotypet for
å finne homozygote linjer. 20 homozygote linjer be identifisert og representerer 17 av CWI
kandidatgenene. Et nytt Acinetobacter sp. ADPWH_lux salisylsyre (SA) biosensor assay ble
brukt til å undersøke om insersjonene påvirker isoxaben-indusert SA produksjon. Insersjon i
fem kandidatgener (TAX2, ZAT11, ZAT18, PRX56 and DUF567 (AT5G41590)) så ut til å
resultere i lavere isoxaben-indusert SA nivå enn villtype kontroll. SA fenotypene som ble
observert med bioassayet ble validert med LC-MS/MS for tre T-DNA insertjonslinjer, tax2,
zat11 and zat18. Dessuten ble de tidligere transkriptomikkdataene for tre av genene (TAX2,
ZAT11 and ZAT18) bekreftet med kvantitativ RT-PCR. Ytterligere qRT-PCR-basert analyse av
genuttrykk indikerte at zat11 trolig er en overuttrykkslinje mens zat18 ble bekreftet som en
knock-out linje. Analyse av en ZAT18::GUS reporter antyder at ZAT18 er høyt uttrykt i
voksende vev som respons på isoxaben-indusert cellulosesyntese inhibering. TAX2 koder for et
antatt signaleringspeptid mens ZAT11 og 18 koder for antatte transkripsjonsregulatorer. De
observerte fenotypene antyder at alle tre genene er involvert i CWIM som respons på
celleveggskader (CWD). Resultatene indikerer at SA bioassayet er et nyttig verktøy for
indentifisering av nye komponenter av CWIM på en effektiv og pålitelig måte. De tre
kandidatgenene som ble mer detaljert karakterisert virker som de spesifikt kreves for regulering
av CWD-indusert SA produksjon. Disse resultatene kan skape muligheter for å dissekere
mekanismene som er ansvarlige for induksjon og modulering av SA som respons på CWD. | |
dc.description.abstract | The plant cell wall provides structural support and protects the cells against environmental
stress. The cell wall integrity (CWI) is maintained through a dedicated CWI maintenance
(CWIM) mechanism and better understanding of this process could aid in the improvement of
food and bioenergy crop performance. Here, 30 T-DNA insertion lines for 24 candidate genes
implicated in CWI based on previous transcriptomics analysis, were genotyped to find
homozygous lines. 20 homozygous lines were identified, representing 17 of the CWI candidate
genes. A novel Acinetobacter sp. ADPWH_lux salicylic acid (SA) biosensor assay was used to
investigate if the insertions affect isoxaben-induced SA production. Insertions in five candidate
genes (TAX2, ZAT11, ZAT18, PRX56 and DUF567 (AT5G41590)) seemed to result in lower
isoxaben-induced SA levels than in wild-type control seedlings. The SA phenotypes observed
with the bioassay were validated with LC-MS/MS for three T-DNA insertion lines, tax2, zat11
and zat18. Furthermore, the preliminary transcriptomics data for three of the genes (TAX2,
ZAT11 and ZAT18) was confirmed by quantitative RT-PCR. Additional qRT-PCR-based
expression analysis indicated that zat11 is likely an overexpression line, while confirming that
zat18 is a knockout line. Analysis of a ZAT18::GUS reporter construct suggests that ZAT18 is
highly expressed in growing tissue in response to isoxaben-induced cellulose synthesis
inhibition. TAX2 encodes a putative signalling peptide while ZAT11 and 18 encode putative
transcriptional regulators. The phenotypes observed implicate all three genes in CWIM in
response to cell wall damage (CWD). The results indicate that the SA bioassay is a useful tool
for identification of novel components of CWIM in an efficient and reliable manner. The three
candidate genes characterized in more detail seem to be specifically required for regulation of
CWD-induced SA production. These results may generate opportunities to dissect mechanisms
responsible for SA induction and modulation in response to CWD. | |
