dc.description.abstract | Bakgrunn: For pasienter med diabetes type 1 er det en daglig kamp for å oppnå god glukose
styre. Dårlig glukosekontroll over tid øker risikoen for mikro- og makrovaskulær
komplikasjoner. Flere subkutane injeksjoner med insulin er den vanligste behandlingen.
Det er flere ulemper med subkutan (SC) administrasjon av insulin og dagens
Hurtigvirkende insulinanaloger stemmer ikke overens med blodglukoseprofilen etter et måltid. en
kunstig pankreas som administrerer insulin intraperitonealt (IP) basert på kontinuerlig
glukose målinger vil hjelpe pasienter oppnå bedre glukose kontroll. For å opprette en
robust kunstig bukspyttkjertel, er det uoppdaget behov for raskere insulinformulering med
Forbedret absorpsjonshastighet, kortere varighet enn de som er tilgjengelige.

dc.description.abstract | Background: For patients with diabetes type 1 it’s a daily struggle to achieve good glucose
control. Poor glucose control over time increases the risk of micro- and macrovascular
complications. Multiple subcutaneous injections with insulin is the most common treatment.
There are several drawbacks with subcutaneous (SC) administration of insulin and the current
rapid acting insulin analogues doesn’t match the blood glucose profile after a meal. An
artificial pancreas which administers insulin intraperitoneally (IP) based on continuous
glucose measurements will help patients achieve better glucose control. In order to create a
robust artificial pancreas, there is an unmet need for a faster insulin formulation with
improved absorption rate, shorter duration of action than those who are currently available.
Method: A search in Embase was preformed to investigate what has been done to improve
the absorption rate of insulin.
Results: The main strategies to increase the absorption rate of insulin are, loosening of tight
junctions, alterations of the extracellular matrix, increase blood flow at the site of
administration, increase the dissociation rate of insulin hexamers and increase the
permeability of endothelial cells by altering the cell membrane.
Conclusion: Increase local blood flow and dissociation rate of insulin hexamers seems to be
the most promising way of enhancing the absorption rate of insulin. | |
