Optimalisering av en protokoll for analyse av cellesyklus i Chinook-laksembryonceller (CHSE-214) ved bruk av flytcytometri: Analyse av cellecyklus som respons på termisk stress

Abstract

Denne rapporten presenterer prosessen av optimaliseringen av en protokoll for studiet av celle syklusen i Chinook laks embryonale celler (CHSE-214) ved bruk av flow cytometri. Studien resulterte i produksjon av en optimalisert metode for undersøkelsen av cellesyklusen i CHSE-214-celler ved bruk av en PI/RNase-fargeløsning.

Denne rapporten presenterer også en studie av de teoretiske og praktiske syn på effekten av temperatur på CHSE-214 celler ved å analysere utviklingen og variasjonene av cellecyklus. For studien ble cellene dyrket ved 20 ° C (CHSE-214 kontroll) og ved 26 ° C (CHSE-214 stresset) og farget med propidiumjodid (PI) for ytterligere proliferasjonsanalyse ved bruk av flow cytometri. This report presents the process of the optimisation of a protocol for the study of the cell cycle in Chinook salmon embryonic cells (CHSE-214) using flow cytometry. The study resulted in the production of an optimised method for the study of the cell cycle in CHSE-214 cells using a PI/RNase staining solution.

This report also presents a study of the theoretical and practical views of the effect of temperature on CHSE-214 cells by analysing their cell cycle development and variations. For the study, the cells were cultivated at 20 °C (CHSE-214 control) and at 26 °C (CHSE-214 stressed) and stained with propidium iodide (PI) for further proliferation analysis using flow cytometry. Conclusions on the results could not be drawn due to the low amounts of parallels used in the study.
